Objective:The purpose of this study was to detect the expression of Rudhira / BCAS3 and Cdc42 in the villus tissue of patients with early embryo cessation,and analyze the relationship between the two indexs,and exploring possible mechanism of embryo cessation,provide a theoretical basis for the prevention and treatment of embryo cessation.Methods:1.Collected 40 patients who had undergone abortion at the Obstetrics and Gynecology Clinic of the Second Hospital of Shanxi Medical University from March to August 2019.Among them,20 patients had abortions due to embryo cessation(experimental group)and 20 cases of early normal pregnancy due to voluntary termination of pregnancy(control group);2.Real-time fluorescent quantitative PCR was used to detect the expression levels of Rudhira / BCAS3 and Cdc42 mRNA in the villi of each group;3.The immunohistochemical SP method was used to detect the expression of Rudhira /BCAS3 and Cdc42 protein in the villous tissue of each group;4.Statistical methods were analyzed using SPSS 22.0 software.All data were tested fornormality and homogeneity of variance.Non-normally distributed measurement data were subjected to natural logarithmic transformation before statistical analysis.The mean plus or minus the standard deviation is expressed;the comparison between the two groups is by t test,and the correlation between the two variables using Pearson analysis.P <0.05 is considered to be statistically significant.Results:1.qRT-PCR test results and correlation1.1 The relative expression of Rudhira / BCAS3 mRNA of the villous tissue in the experimental group was 1.52 ± 1.28 and 2.76 ± 1.84 in the control group.The level of Rudhira / BCAS3 mRNA of the villous tissue was lower in the experimental group,and the difference was significant(P < 0.05);1.2 The relative expression of Cdc42 mRNA of the villous tissue in the experimental group was 1.17 ± 1.23 and 2.61 ± 1.27 in the control group.Compare to the control group,the expression of Cdc42 mRNA of the villous tissue in the experimental group was lower than the control group,and the difference was significant(P <0.05);1.3 Pearson correlation analysis of the results of RT-PCR experiments showed that the correlation between Rudhira / BCAS3 mRNA and Cdc42 mRNA of the villi tissue in the experimental group was significant positive(r=0.57,P=0.009).2.Immunohistochemical SP test results2.1 Rudhira / BCAS3 protein is mainly expressed at the cell membrane and cytoplasm.The Rudhira / BCAS3 staining score of the villous tissue in the experimental group and the control group were 2.55 ± 0.69 and 4.65 ± 0.81 separately.Compare to the control group,the Rudhira / BCAS3 staining score of the villous tissue in the experimental group was lower.The difference was statistically significant(P <0.05);2.2 Cdc42 protein is mainly distributed on the cell membrane and cytoplasm.The Cdc42 staining score of the villous tissue in the experimental group was 1.75 ± 0.72,and the control group was 4.45 ± 0.99.The Cdc42 staining score of the villous tissue in the experimental group was lower than in the control group.(P <0.05).Conclusion:Compare to the control group,The level of Rudhira / BCAS3 mRNA and Cdc42 mRNA of the villus tissue in the experimental group was lower,and also the expression of Rudhira / BCAS3 protein and Cdc42 protein of the villus tissue in the experimental group was lower;There was a positive correlation between Rudhira /BCAS3 mRNA and Cdc42 mRNA expression of the villi in the experimental group.
