Objective:The expression of HIF-1a,VEGF and Cathepsin S in embryonic suspension and normal early pregnancy were studied,and their possible roles and interconnections in embryogenesis mechanism were explored.Methods:Grouping(1)Embryo Stopping Group: From March 2018 to August 2018,20 cases of abortion were confirmed by B-ultrasound in our outpatient clinic.(2)Normal early pregnancy group: 20 cases of early pregnancy with artificial abortion were selected in the outpatient clinic because of the unsuccessful pregnancy.Inclusion criteria: 49-70 days after menopause,confirmed by intrauterine pregnancy by B-ultrasound,but the embryo has no heart tube beat and stopped development,no hormone drugs have been taken in the past 3 months;blood routine,coagulation function is normal;leucorrhea routine no obvious abnormalities;History of reproductive anatomy and history of genital tract infection;no history of infectious diseases;both sides denied the history of genetic disease;chromosomal gene detection excluded the chromosomal problems caused by embryo cessation,specimens were collected and approved by the Ethics Committee of the Second Hospital of Shanxi Medical University,patients Both informed and signed the consent form,and the research method met the ethical standards.Exclusion criteria: genital malformations;suffering from immune system diseases;infectious diseases in the first 3 months of pregnancy,use of immunosuppressive agents;combined with endocrine abnormalities;recent history of surgical trauma.The expression levels of HIF-1a,VEGF and Cathepsin S mRNA in embryonic suspension group were detected by RT-PCR.The expression of HIF-1a,VEGF and Cathepsin S in normal early pregnancy group was detected by the same method.The study used SPSS20.0 software for statistical analysis.All the data were tested for normality and homogeneity of variance.The non-normally distributed measurement data were statistically analyzed after natural logarithmic transformation.The comparison between the two groups was selected.For the t test,the correlation between the indicators was analyzed by Pearson analysis,and P < 0.05 was considered to be statistically significant.Results:There was no significant difference in the general data between the embryonic suspension group and the normal early pregnancy group,ie age and gestational age(P>0.05).The results of RT-PCR showed that the relative expressions of HIF-1a,VEGF and CathepinS in the embryonic suspension group were lower than those in the normal early pregnancy group,and the indexes of embryonic suspension group and normal early pregnancy group were tested by independent sample t test,P<0.05.There was a statistically significant difference in the expression of HIF-1a,VEGF and Cathepsin S between the embryonic suspension group and the normal early pregnancy group.There was a positive correlation between HIF-1a and VEGF,VEGF and Cathepsin S by Pearson correlation analysis.And P < 0.05,the difference was statistically significant.Conclusion:HIF-1a,VEGF and Cathepsin S decreased in embryonic suspension,which may be involved in embryonic cessation,resulting in defects and dysfunction ofembryonic vascular formation,which further led to embryonic blood supply,inadequate feeding and nutrient deficiencies.Make the embryo go to a bad pregnancy outcome-embryos are stopped.
