Abstract/Summary:
PurposeThis study investigated the expression levels of Treg/Th17-related factors in embryo transfer,natural pregnancy abortion patients and normal early pregnant women;and explored the differences and correlations between expression in abortion patients and normal early pregnant women.MethodThirty patients who underwent in vitro fertilization-embryo transfer(IVF-ET)abortion from the Reproductive Center of Lanzhou First Hospital from January 2018 to August 2018 were selected as observation group(group A),and family planning clinic in Gansu provincial people’s Hospital.Sixty patients underwent artificial abortion,of which 30 were aborted in the observation group(group B),and 30 normal pregnant women were in the control group(group C).The levels of IL-6,IL-10 and IL-17 cytokines in serum were determined by enzyme-linked immunosorbent assay(ELISA).The expression levels of IL-6,IL-10,IL-17 and Foxp3 m RNA in villus were detected by RT-PCR.Compare the characteristics and differences between the three groups.ResultThere was no significant difference in the age,gestational age,menstrual cycle and body mass index of the patients included in the experiment(P>0.05).2.The results of ELISA showed that the levels of IL-10 in group A and group B were lower than those in group C,while the levels of IL-6 and IL-17 were increased,the difference was statistically significant(P<0.05).There was no significant difference in the level of difference between group A and group B(P>0.05).3.RT-PCR results showed that IL-6,IL-10,IL-17,and Foxp3 were expressed in the villus of the three groups.The expression of IL-17 in the villius of group A and group B was higher than that of group C,while the expressions of IL-6,IL-10 and Foxp3 m RNA were lower than those in group C.There was no significant difference in the expression levels between group A and group B(P>0.05).Conclusion1.The expression of inflammatory factor IL-17 in serum and chorionic villi of patients with embryonic sterilization was significantly higher than that of normal early pregnant women,suggesting that abortion may occur by inducing aggregation of inflammatory factors at the maternal-fetal interface.2.The concentration of IL-6 in serum of patients with embryo arrest was higher than that of normal early pregnant women,but the m RNA expression level in group was decreased,suggesting the imbalance of IL-6 level and the imbalance and promotion of immune tolerance and anti-inflammatory effects.Embryo implantation and pregnancy maintenance are related.3.The expression of Treg cell related factor IL-10,Foxp3 in serum and chorionic villi of patients with embryonic sterilization was significantly lower than that of normal early pregnant women,suggesting that the Treg cytokine-mediated immune response may be enhanced to the abnormal implantation of trophoblast cells,leading to miscarriage.4.There was no significant difference in the expression of cytokines in serum and villi between the two groups(P > 0.05).The results showed that there was no significant difference in immune level in vivo during embryo sterilization with different modes of conception.
