Objective:By studying the expression of HIF-2α,VEGF and SOX7 in villus tissue of patients with normal early pregnancy and early missed abortion,and then analyze the relationship and significance of the three in the pathogenesis of missed abortion.Methods:1.Collecting 50 cases who received induced abortion in the Obstetrics and Gynecology Abortion Room of Our hospital from August 2021 to January 2022,including 25 patients who underwent induced abortion due to early missed abortion confirmed by B ultrasound(early missed abortion group)and 25 women who were confirmed by B ultrasound as intrauterine early pregnancy and voluntarily underwent induced abortion(normal early pregnancy group).2.Useing immunohistochemical method to check the expression of HIF-2α,VEGF and SOX7 in villus tissues of the control group;using the technique of Western blot to measure the expression changes of HIF-2α,VEGF and SOX7 protein in the above tissues,and the relationship between the three in missed abortion was analyzed.3.Statistical analysis was performed with SPSS 25.0 software.First,testing experimental data with normality and variance homogeneity,and if they did not meet the normality,the data should be log-transformed before analysis.The comparison of the same index was performed by t-test,then the relevancy between variables in one same group was analyzed by the method of Pearson correlation analysis.P < 0.05 was considered statistically significant.Results:1.There was no statistical significance in the comparison of the general information of the early missed abortion group and the normal early pregnancy group,including age,number of pregnancies,blood pressure,etc(P>0.05).2.Immunohistochemical results The expression areas of HIF-2α,VEGF,and SOX7 in the villus tissues of the early missed abortion group were larger than those of the normal early pregnancy group,and the mean optical density values(AOD values)were compared using Image J software.Quantitative analysis of the results showed that the expression levels of HIF-2α,VEGF,and SOX7 in the villus tissues of the early missed abortion group were reduced compared with those of the normal early pregnancy group,and the difference between them was statistically significant(P < 0.05).3.Western Blot Results The expression levels of HIF-2α,VEGF and SOX7 in early missed abortion(2359.05 ± 608.72,1548.66 ± 580.82,2359.05 ± 608.72,respectively)were lower than those in normal early pregnancy(7261.81 ± 927.26,7352.35 ± 911.10,6990.57 ± 990.74,respectively),with statistical significance(P < 0.05);Pearson correlation analysis revealed a positive correlation between the three indicators,with statistical significance(P < 0.05).Conclusion:1.The expression of HIF-2α,VEGF and SOX7 in villus tissue of the two groups.2.The expression of HIF-2α,VEGF and SOX7 protein in villus tissue of the early missed abortion group was significantly lower than that of the normal early pregnancy group.3.There was a positive correlation between HIF-2α,VEGF and SOX7 in villus tissue of early missed abortion group,suggesting that the low expression of the three is involved in the pathogenesis of early missed abortion,resulting in poor embryonic angiogenesis,which in turn leads to adverse pregnancy outcomes-early missed abortion. |
