Objective:1.To observe the effects of MBWP on the learning and memory function of the APP/PS1 transgenic mice.2.To observe the effects of MBWP on NF-?B and related inflammatory factors in hippocampus of APP/PS1 transgenic mice.3.To investigate the mechanism of MBWP in improving the learning and memory ability of APP/PS1 transgenic mice.Methods:1.Thirty APP/PS1 transgenic mice were divided into three groups on average:APP/PS1 group,low-dose MBWP group(L-MBWP),high-dose MBWP(H-MBWP)group.Ten healthy mice served as a healthy control group.2.The healthy control group and APP/PS1 group were treated with normal saline(1ml/100 g body weight)by gavage;the L-MBWP and H-MBWP groups were given gavage treatment with low-dose MBWP(0.15g/ml,1ml/100 g body weight)and high-dose MBWP(0.30g/ml,1ml/100 g body weight)respectively,once a day for 4months.3.Spontaneous alternation behavior in a Y-maze were recorded to evaluate the spatial learning and memory abilities of four groups mice.4.Western blot was used to detect the expression levels of NF-?B and TNF-?protein in the hippocampus of four groups mice.5.Statistical analysis was performed using Graph Pad Prism 7.0 software.The measurement data is expressed as mean±standard deviation(x±s).The comparison of differences between multiple groups is tested by single factor analysis of variance,and the comparison of groups is by LSD-t test line.?= 0.05 is the inspection level,and P<0.05 is regarded as statistically significant.Results:1.The effects of MBWP on the learning and memory function of the APP/PS1 transgenic mice: The percentage of alternations in the Y-maze test was significantly lower in the APP/PS1 group than in the control group(P<0.01).In addition,the percentage of alternations was significantly higher in the H-MBWP group than in the APP/PS1 group(P<0.01).Compared with APP/PS1 group,there was no significant difference in L-MBWP group(P>0.05).However,the total number of arm entries did not significantly differ among groups.2.The effects of MBWP on the expression of NF-?B p65 in hippocampus of APP/PS1 transgenic mice.The expression of NF-?B p65 was significantly increased in the APP/PS1 groups compared to that in the control group(P<0.01).In addition,NF-?B expression was significantly reduced in the H-MBWP group compared to that in the APP/PS1 group(P<0.05).Although NF-?B p65 expression was reduced in the L-MBWP group compared to that in the APP/PS1 group,the difference failed to reach significance(P>0.05).3.The effects of MBWP on the expression of TNF-? in hippocampus of APP/PS1 transgenic mice.The TNF? level was significantly increased in the APP/PS1 and L-MBWP groups compared to that in the control group(P<0.01,P<0.05,respectively).In addition,the TNF? level was significantly reduced in the H-MBWP group compared to that in the APP/PS1 group(P<0.01).Compared with APP/PS1 group,there was no significant difference in L-MBWP group(P>0.05).Conclusion:1.MBWP can improve the learning and memory ability of AD APP/PS1 transgenic mice.2.The expression of NF-?B p65 and related inflammatory factor TNF-? in hippocampus of APP/PS1 transgenic mice are increased.3.MBWP can inhibit the activation of NF-?B p65 and the release of inflammatory factor TNF-? in the hippocampal tissues of APP/PS1 transgenic mice,suggesting that this compound might play a neuroprotective role by inhibiting the neuroinflammation in hippocampi.
