Objective:1.To explore the effects of modified bu wang powder on the the pathologic changes and apoptosis in the hippocampal neuron of AD model rats;2.To observe the effect of modified bu wang powder on the neuroinflammation in hippocampus of AD model rats.Methods:1.32 male SD rats of 8 weeks.Equally separated into control group,AD model group,A?+low dose treatment of modified bu wang powder group and A?+high dose of modified bu wang powder treatment group,8 for each group;2.To establish AD rat model by using the brain stereotaxic instrument to inject A?1-42 into bilateral hippocampus;3.To examine the learning and memory of the rats by Y maze;4.To explore the pathologic changes of nerve cells in the hippocampus by HE;5.To examine the apoptosis of nerve cells in the hippocampus by TUNEL;6.Detection of inflammatory factor levels in hippocampus of rats by enzyme linked immunosorbent assay(ELISA).Results:1.The effect of modified bu wang powder on learning and memory ability of AD model rats.Results of Y maze test: there was no considerable distinction in the whole number of arm entries in the four groups;spontaneous alternation ratio results: contrast to the control group,AD model group has decreased spontaneous alternation(P<0.05),there was no distinction among the A?+low dose treatment of modified bu wang powder group and the AD model group(P>0.05),while it was notably increased in the A?+high dose of modified bu wang powder treatment group than the AD model group(P<0.05).2.The effect of modified bu wang powder on morphological effects in hippocampal neurons of AD model rats.The outcome of HE demonstrated that the nerve cells in the hippocampus in the control group had normal morphology,intact structure,neatly arranged,and no necrotic neurons were found.The hippocampal neurons in the AD model group had aberrant morphology,loose structure and disrupted arrangement,pyknotic nuclei can also be expressed.The A?+low dose treatment of modified bu wang powder group was similar to the AD model group,with the disordered cell arrangement,and there were still necrotic neurons.Contrast to model group,the quantity of neurons in the A?+high dose of modified bu wang powder treatment group was significantly increased,and the cell structure was relatively intact and neat arrangement.3.The effect of modified bu wang powder on apoptosis of nerve cells in the hippocampus of AD model rats.Results of TUNEL staining: the amount of apoptotic cells in control group was extremely few.There were a quantity of apoptotic cells in AD model group contrast to control group(P<0.0001).There existed no distinction in the amount of apoptotic cells among A?+low dose treatment of modified bu wang powder group and AD model group(P>0.05),but the amount of apoptotic cells in A?+high dose of modified bu wang powder treatment group was obviously reduced than AD model group(P=0.005).4.The effect of modified bu wang powder on neuroinflammation in hippocampus of AD model rats.The level of IL-1? in hippocampus: it was increased in AD model group contrast to control group(P<0.0001),and the A?+low dose treatment of modified bu wang powder group was not lower than the AD model group(P=0.98),A?+high dose of modified bu wang powder treatment group was lower than the AD model group(P<0.001).The level of IL-6 in hippocampus: The AD model group was greater in amount than control group(P<0.0001),A?+low dose treatment of modified bu wang powder group was not lower than AD model group(P=0.99),however,A?+high dose of modified bu wang powder treatment group was lower than AD model group(P=0.01).The level of TNF-? in hippocampus: compared with the control group,it was larger in amount in the AD model group(P<0.0001).The level of TNF-? in hippocampus in A?+low dose treatment of modified bu wang powder group was not greater in amount than the AD model group(P=0.26),but it was reduced in A?+high dose of modified bu wang powder treatment group(P<0.0001).5.The relationship between learning and memory ability and neuroinflammation and neuronal apoptosis in ratsThe correlation analysis showed that the correlation coefficient between spontaneous rate of Y maze and TUNEL positive rate and the levels of inflammatory factors IL-1?,IL-6 and TNF-? in hippocampus were-0.77,-0.81,-0.60,-0.68,respectively(P<0.05).The correlation coefficient between TUNEL positive rate and hippocampal inflammatory factors IL-1?,IL-6 and TNF-? levels was 0.77,0.63,and 0.63,respectively(P<0.05).Conclusions:1.Modified bu wang powder can enhance the learning and memory ability of AD model rats.2.Modified bu wang powder ameliorate the morphology of hippocampal neurons of AD model rats better.3.Modified bu wang powder could inhibit the hippocampal neuroinflammation and the apoptosis of nerve cells induced by A?1-42.4.The learning and memory ability of rats increased with the decrease of neuronal apoptosis and neuroinflammatory,and the more severe the apoptosis of rat hippocampal neurons,the more serious the degree of neuroinflammatory.
