Objective: To investigate whether exogenous hydrogen sulfide(H2S)can regulate the content of miRNA-455 in rat epidermal cells under the intervention of burn serum and The content of miRNA-455 in serum cells of burned rats.Methods: Rat and rat epidermal cells were studied.Animal level: 20 male SD rats weighing 350-400 g were selected.They were randomly divided into 3 groups: normal control group,burn group and burn + Na HS group.In the burn group and the burn + Na HS group,a III° burn model with a burn area of 30% was made on the back after anesthesia.The burn + Na HS group,56 ?mol /L of Na HS solution 0.4ml/100 g is injected intraperitoneally 1d and 2d after the burn,respectively.The normal control group,the burn group and the burn + Na HS group was fed under the same conditions.After burns for 2d and 3d,blood was collected from the abdominal aorta to prepare the serum.The total RNA in the serum of each group was extracted and the content of miRNA-455 in each group was detected by q PCR.Cell level:rat epidermal cells was randomly divided into 3 groups: normal group,burn serum control group,and burn serum +Na HS group.After synchronous culture of the 3 groups of cells,the 3 groups of cells were inoculated into a 6-well plate for culture.The normal group continued to use fetal bovine serum culture.In burn serum control group and burn serum +Na HS group,10% burn rat serum medium was used to intervene for 4h.After The burn serum +Na HS group was treated with Na HS solution with a concentration of 150?mol / L,at 1h?4h and 24 h,respectively.Total RNA was extracted from epidermal cells in each group and the content of miRNA-455 in each group was detected by q PCR.Results: 1.At the same time point.Animal level: The difference between the normal group and the burn group is statistically significant at 2d(P < 0.05),and the expression level of miRNA-455 in the burn group is significantly higher than that in the normal group.Cell level: the difference between the normal group and the burn serum group was statistically significant after 1h(P < 0.05),and the expression level of miRNA-455 in the burn serum control group is significantly higher than that in the normal group.2.At the same time point.Animal level: the expression level of miRNA-455 in the burn + Na HS group is significantly lower than that in the burn group(P < 0.05),and the difference is statistically significant.Cell level: compared with the burn + Na HS group,the expression level of miRNA-455 in the burn + Na HS group is significantly lower than that in the burn + Na HS group(P < 0.05),and the difference is statistically significant.Conclusions: 1.The content of miRNA-455 in burned rats is significantly higher than that in normal rats.2.Exogenous hydrogen sulfide Na HS at a concentration of 150?mol / L,can obviously reduce miRNA-455 content in serum cells of burned rats.3.Exogenous hydrogen sulfide Na HS at a concentration of 56 ?mol / L,can obviously reduce miRNA-455 content in the epidermal cells of burned rats.
