Objective To investigate the impact of hydrogen sulfide on endoplasmic reticulum stress in Burned Rats Serum Intervention Cultured Epidermal Cells Methods The epidermal cells extract from the back epidermis of 1 day old neonatal rats were selected as research objects.The research group was divided into three groups.Choose 52 healthy SD rats(half male and female),normal feeding to 350-400 g then keep them 24 hours of fasting,randomly select 10 healthy SD rats(half male and female)for taking blood serum from aorta abdominalisunder with sterile condition,the blood serum taking from centrifugal extraction used as normal control group for standby;the blood serum taking from 42 SD rats(half male and female)that through 18 hours burn molding treatment used as the experimental group.Took the experimental group randomly divided into two groups,used as the burn control group and burn intervention group(exogenous NaHS intervention group).In the process of cultivation,the normal control group and the burn control group were prepared into 10% normal and 10% burn rat serum medium respectively,and the burn intervention group were prepared as 10% burn rats serum medium,then inject with NaHS with concentration of 200?mol/L.Epidermal cells were cultured in vitro for 1 hour,6 hours,12 hours and 24 hours respectively.After cell culture for 1 hour and 6 hours and 12 hours,RT-PCR was used to detect the relative mRNA expression levels of endoplasmic reticulum stress of JNK,glucose-regulated protein 78(GRP78),calreticulin(calreticulin,CRT)and caspase-12 in epidermal cells.After 24 hours cell culture,Western Blotting blot assay was used to detect the protein expression level of phosphoric acid JNK,GRP78,CRT and caspase-12.Results At each different time point,the mRNA expression levels of endoplasmic reticulum stress of JNK,GRP78,CRT and caspase-12 in epidermal cells of burn control group were significantly higher than those in normal control group at the same time point and the difference were statistically significant(P < 0.05).Similarly,at the same time point,the mRNA expression levels of JNK,GRP78,CRT and caspase-12 in epidermal cells of burn intervention group were significantly lower than those in burn control group,the difference were statistically significant(P < 0.05).Meanwhile,at the same after-24-hours time point,the protein expression level of endoplasmic reticulum stress of phosphoric acid JNK,GRP78,CRT and caspase-12 in epidermal cells of burn control group were significantly higher than those of normal control group at each time point,the difference was statistically significant(P < 0.05).At the same time point,the protein expression level of endoplasmic reticulum stress of phosphoric acid JNK,GRP78,CRT and caspase-12 in epidermal cells of burn intervention group were significantly lower than those of burn control group,the difference was statistically significant(P < 0.05).Conclusion As a stress injury factor,burn can increase the expression level of endoplasmic reticulum stress in epidermal cells,and a small amount of exogenous hydrogen sulfide can reduce the expression level of endoplasmic reticulum stress in epidermal cells,alleviate the effect of endoplasmic reticulum stress on cell injury,reduce the level of apoptosis after burn,and play a certain role in wound protection.
