Objective To investigate the effect of exogenous hydrogen sulfide on calcium ion and calmodulin concentration of epidermal cells in burn serum.Methods 350g-400 g healthy male SD rats were selected to establish 30% TBSA III degree burn rat models.Blood samples were collected from the abdominal aorta of normal rats and burned rats to prepare normal serum and burn serum for use.Third generation rat epidermal cells were cultured in vitro.Normal serum,burn serum and burn serum with Na SH were used to intervene epidermal cells respectively.Cells in each group were collected.Fluorescent staining of intracellular calcium ions by Fluo-4,AM was performed and the fluorescence intensity of intracellular calcium ions was detected by flow cytometry;The expression level of calmodulin was detected by western blot.Results(1)The average fluorescence intensity of calcium ions in the treatment group of normal serum intervention epidermal cell(12177.11±478.77)was significantly lower than that in the treatment group of burn serum intervention epidermal cell(14852.33±313.60)(p<0.05).(2)The average fluorescence intensity of calcium ions in the treatment group of burn serum intervention epidermal cell(14852.33±313.60)was significantly higher than that in the treatment group of burn serum with sodium hydrosulfide intervention epidermal cell for 1.0h(12325.00±286.36)(p<0.05).(3)The average fluorescence intensity of calcium ions in the treatment group of burn serum with sodium hydrosulfide intervention epidermal cell for 1.0h(12325.00±286.36)was significantly lower than that in the treatment group of burn serum with sodium hydrosulfide intervention epidermal cell for 0.5h(14467.33±621.20)(p<0.05).(4)The protein expression level of calmodulin in the treatment group of normal serum intervention epidermal cell(1.35±0.39)was lower than that in the treatment group of burn serum intervention epidermal cell(1.78±0.21),but the difference was not statistically significant(p>0.05).(5)The protein expression level of calmodulin in the treatment group of burn serum intervention epidermal cell(1.78±0.21)was significantly lower than that in the treatment group of burn serum with sodium hydrosulfide intervention epidermal cell for 1.0h(2.58±0.44)(p<0.05).Conclusions Exogenous hydrogen sulfide can reduce the unbound calcium ion concentration in epidermal cells after burn stress in a certain time range.Exogenous exogenous hydrogen sulfide can increase the protein expression of calmodulin in epidermal cells after burn stress.
