Objective To evaluate the anti proliferation,migration and invasion effects of Matrine on tu686 cells and tu686 cells transfected with EBV.Methods The laryngeal cancer TU686 cells were obtained by cell culture.The logarithmic growth of laryngeal cancer TU686 cells were divided into the first group(0mg/ml),the second group(0.5 mg/ml),the third group(1 mg/ml),the fourth group(2mg/ml)and the fifth group(4mg/ml),CCK-8 method based on WST-8 colorimetric detection kit was used to determine the activity of laryngeal cancer cells,and the appropriate drug concentration for the next experiment was obtained;after EBV was transfected into laryngeal cancer TU686 cells,the content of EBV-DNA was detected by RT-PCR,and then it was divided into untreated group(control group)and Matrine treatment group(the final concentration of matrine is 1mg/ml),EBV treatment group(the final concentration of EBV is20%(V / V))and matrine + EBV treatment group,The effects of Matrine on the apoptosis,migration and invasion of EBV(+/-)laryngeal carcinoma TU686 cells were detected by flow cytometry,Scratch test and Transwell cell test respectively.The effects of matrine and EBV on the expression of matrix metalloproteinase-2(MMP-2),NF-?Bp65 and NF-?BP50in the cell nucleus of TU686 cells were analyzed by RT-PCR and Western blot.Results1.Matrine can reduce the proliferation of TU686 cells and Hep-2 cells in a dose-dependent manner.The inhibition rates of 24-hour matrine on TU686 cells were 47.37%,65.87%,83.48% and 91.92%(P<0.05).matrine could inhibit the migration and invasion ofTU686 cells,promote cell apoptosis,reduce the expression level of matrix(MMP-2)RNA and protein,and significantly reduce the expression level of NF-?Bp50,NF-?Bp65 RNA and protein in TU686 cells.2.Matrine could inhibit the ability of migrate and invade,which laryngeal cancer TU686 cells transfected with EBV,matrine could promote laryngeal cancer TU686 transfected with EBV apoptosis of cells,also could inhibition of metalloproteinases in cells-2(MMP-2),NF-?B p50,NF-?B p65 RNA and protein expression.3.But the results of EBV after intervention were just the opposite.Conclusion Matrine has the ability to inhibit the growth and metastasis of TU686 cells and TU686 cells transfected with EBV.
