Objective:To investigate the effect of folic acid on autophagy-related proteins of tumor tissue cells in mice with cervical cancer.Methods:1?SiHa cell lines of related cervical cancer cells were cultured in vitro,and cells in the logarithmic growth stage were taken to establish the transplanted tumor model of cervical cancer in nude mice.2?The nude mice were randomly divided into 3 groups:(1)folate deficiency group(folate deficiency feed + distilled water gavage)(2)folate supplementation group(normal feed + folic acid 600ug/kg/d gavage)(3)normal control group(normal feed + distilled water gavage).The body weight was weighed once a week,and the dosage of gavage was adjusted according to the body weight.3?Western blot was used to detect the expression changes of autophagy-related proteins LC3 and p62 in transplanted tumor cells of cervical cancer in nude mice at the protein level.Fluorescence quantitative PCR was used to detect the expression changes of autophagy-related proteins LC3 and p62 in transplanted tumor cells of cervical cancer in nude mice at mRNA level.Results:1?In the transplanted tumor model of cervical cancer in nude mice,the expression of autophagy-related protein LC3 in transplanted tumor cells in the folate supplementation group was lower than that in the control group(1.0285±0.8623)and the folate deficiency group(1.4856±0.0823),and the expression of autophagy-related protein LC3 in the folate deficiency group was higher than that in the control group.The expression ofautophagy-related protein P62 was(2.037±0.0605)higher in the folate supplementation group than in the control group(1.7302±0.0474)and the folate deficiency group(1.0410±0.0311).The content of autophagy-related protein P62 in the folate deficiency group was lower than that in the control group(P < 0.05,the difference was statistically significant).2?Compared with the control group,LC3 expression was higher and P62 expression was lower in the folate deficiency group and the folate supplement group(P < 0.05).Conclusion:1?Compared with the control group,the expression of autophagy-related protein LC3 was high and the expression of P62 was low in the folate deficient group,while the expression of autophagy-related protein LC3 was low and the expression of P62 was high in the folate supplementation group.2?Folic acid deficiency can promote the expression of autophagy-related protein LC3 in transplanted tumor cells of cervical cancer nude mice,and promote the improvement of autophagy level in transplanted tumor cells of cervical cancer nude mice.3?Compared with the folic acid deficient group,the expression of autophagy-related protein LC3 in transplanted tumor cells of nude mice of cervical cancer decreased and the expression of P62 increased,indicating that different doses of folic acid had certain influence on the autophagy level of transplanted tumor cells of nude mice of cervical cancer.
