Objective:Cervical cancer is one of the most common gynecologic malignancies in the world.The incidence and mortality rate of cervical cancer is increasing year by year. Studies have indicated that the treatment of folic acid has a positive significance for the prevention of cervical cancer. Folic acid is a necessary trace element in the body, and epidemiological evidence also suggests that folic acid is closely related to the occurrence of cervical cancer.But there is no clear study on the specific mechanism of action between the two.Autophagy is a unique way of cell death, and autophagy plays a role in the fight against tumor formation. Autophagy by of damaged organelles or abnormal accumulation of materials removal to adjust the cell itself smooth it is beneficial to genome stability,inhibition of transformed cells into cancer cells, resulting in reduced cell cancerization. But the study of the relationship between the three cervical cancer, autophagy and folic acid has not yet been involved, so their relationship is not clear. This study aimed to investigate the effects of folic acid on the expression of autophagy protein βeclin-1 and cervical cancer cell SiHa cell biology function.Methods:1. SiHa cells were cultured under normal conditions, and 1ug/mL(control group),10ug/ml, 100ug/ml, 1000ug/ml and 500ug/ml were used to observe and record the morphological changes and growth rate of cell.2.The use of 1ug / mL(control group), 10 ug / ml, 100 ug / ml, 500 ug / ml, 1000 ug / mlof folic acid intervention. The proliferation activity of SiHa cells of cervical cancer was detected by CCK8. The migration ability of cells was observed by scratch test. Cell apoptosis was analyzed by flow cytometry. Western Bloting was used to detect the expression level of autophagy protein βeclin-1.Results:1.Compared with the control group, folic acid concentration reached 1000ug/ml when cells of morphology change significantly, when folic acid concentration in the experimental group compared to the control group in 100 ug / ml and above, the total number of cells in the experimental group were significantly lower,2.CCK8 cell proliferation in experimental displayed in different concentration of folic acid intervention, in addition to folic acid 10 ug / ml in the experimental group and the control group compared P=0.650 no difference was not statistically significant and, 100ug/ ml, 500ug/ml, 1000ug/ml concentrations of folic acid cell proliferation and control group had statistical significance compared, P = 0.041, 0.001, 0.001, respectively.3.Scratch experiments show that 10 ug/ml the folic acid group of scratches healing area is no obvious difference compared with control group. 100 ug/ml, 500 ug/ml, 1000ug/ml concentration the folic acid group compared with control group of scratches healing area reducing, and with the increase of folic acid concentration healing area reducing increasingly obvious.4. Flow cytometry was used to analysis the effects of folic acid on the apoptosis of cervical cancer cells, the results showed 10 ug / ml of folic acid in the experimental group cells apoptosis rate and the control group compared P=0.580 differences had no statistical significance, the folic acid cells 100 ug / ml, 500ug/ml, 1000ug/ml concentrations of apoptosis rate and control group phase ratio were statistically significant, P = 0.02, 0.001 and 0.001, respectively.5. Wesrern Bloting to detect the effect of folic acid on the expression of autophagy gene βeclin-1 protein in cervical cancer cells. The expression levels of 10ug/ml, 100ug/ml,1000ug/ml and βeclin-1 in the experimental group were significantly different than that in the control group, the difference was statistically significant P values were 0.014, 0.003,0.001, 0.001, respectively.Conclusions:1. High concentration folic acid inhibits the proliferation and migration of cervical cancer SiHa cells, and promotes the apoptosis of SiHa cells.2. High concentrations of folic acid to promote the expression of autophagy proteinβeclin-1 in cervical cancer SiHa cells.
