| Purpose:We explored the expression level of folate receptor alpha(FR-α)in normal cervical tissue,cervical intraepithelial neoplasia(CIN)tissue and cervical cancer tissue to provide cervical cancer and CIN with the new screening and diagnosing method of the scientific evidence.Methods:The 172 patients undergone total hysterectomy were selected from September 2017 to January 2020 in our hospital,including 69 cases of cervical cancer,63cases of CIN(LSIL21 cases,HSIL42 cases)and 40 cases of uterine leiomyoma(as the normal control group),based on the final histopathological examination results.The expression level of FR-αwas tested in different cervical lesions using the immunohistochemistry.Then,8 cases of normal cervix and 8 cases of cervical cancer tissue were randomly selected and the expression level of FR-αwas detected in cervical cancer and normal cervical tissue by qRT-PCR and Western blot.SPSS 25.0 was used for data analysis.Measurement data were expressed as mean±standard deviation.Independent sample T test was used for comparison between groups.Kruskal-wallis single factor ANOVA(K sample)was used for counting data.Chi-square test was used for the difference of positive rate and strong positive rate,and P<0.05 was statistically significant.Results:1)The positive expression of FR-αwas different in normal cervical group,LSIL group,HSIL group and cervical cancer group(Z=25.606,P<0.001).The further analysis subgroups showed that the expression of FR-αwas different between normal control and cervical cancer group or HSIL group(P<0.001).However,there was no difference of the expression of FR-αin normal cervical group and LSIL group,HSIL group and cervical cancer group compared with normal cervical group respectively.2)The positive rate of FR-αexpression in normal cervical group,LSIL group,HSIL group and cervical cancer group was 67.50%,85.70%,88.10%,78.30%respectively,and the difference was not statistically significant(?~2=5.890,P=0.117).The strong positive expression rate of FR-αin normal cervical group,LSIL group,HSIL group and cervical cancer group was 15.0%,33.3%,59.5%,66.7%,respectively,and the difference was statistically significant(?~2=31.048,P<0.001).3)The relative expression level of FR-αmRNA in the cervical cancer group was calculated by considering the normal cervical FR-αmRNA group as the calculation standard.As the result showed that the relative expression of FR-αmRNA between the normal cervical and cervical cancer groups was statistically significant(P=0.005).4)The level of protein expression was tested in normal cervical tissue and cervical cancer tissue using the Western Blot method.And we used polyclonal antibodies against human FR-αand GADPH as the Internal reference genes.A relative gray ratio represents the level of FR-αprotein expression.The results showed that FR-αprotein was highly expressed,the differences was statistically significant(P=0.012)in cervical cancer tissues compared to normal cervical tissues.Conclusion:The expression of FR-αwas decreased in normal cervical tissue,but increased in HSIL and cervical cancer tissue.With the increase of cervical lesion grade,the strong positive expression rate of FR-αincreased gradually,which provide a clue for t screening cervical cancer and HSIL. |
