Objective: To study the sports fatigue in mice IFN-gamma,IL-4,IL-10 and TNF alpha cytokine expression changed,IFN-gamma/IL-4 balance dynamic change characteristics and B7 1,B7-2 gene and protein expression level,further to understand the mechanism of exercise-induced immunosuppression occurs,found delay after the exercise fatigue and to maintain the body movement fatigue immune balance method.Methods:The healthy kunming mice were randomly divided into 5 groups,10 of which were normal control group,the control group of the model,and the group of the drug algari.The mice were trained on the increasing load run.After each exercise group were artificial gastric perfusion 0.2 mL low concentration drug concentration in red,ali dimensions medicine red,high dimensional ali ali diluent,drug doses,respectively for 20,40,80 mg / 100 g weight.Immediately after the last movement,the mice were executed and the materials were taken.Results:(1)mice weight monitoring: compared with M group,the experimental mice weight were higher than in M group,E1 and E2 group compared with M group with significant difference(P < 0.05),E3 and M group is very significant differences(P < 0.01);(2)the mice spleen index and thymus index: experimental mice spleen weight,weight of thymus and spleen index and thymus index compared with group M rise trend,the E1 and M group was no significant difference(P > 0.05),E2 and E3 group compared with M group had significant difference(P < 0.05);(3)the running test of the running table: the time of the running time of the group C was significantly higher than that in group M and E1(P < 0.05).In E2 and E3 groups,there was a significant increase in the exhaustion time(P < 0.05).(4)levels of blood lactate and blood testosterone: the level of blood lactate in M groups was significantly higher than that in group C(P < 0.01)and the experimental groups(P < 0.05).Compared with group C,there was a significant increase in blood lactate level(P < 0.05)in the experimental groups.The serum testosterone levels in group M and E1 group were significantly higher than that in group C(P < 0.01).The E2 group and E3 group were significantly lower than that of group M(P < 0.01),and there was no significant difference between E1 group and M group.(5): the level of serum myocardial enzyme M group of mice serum myocardial enzyme(AST,LDH and CK)were significantly lower than that of group C(P < 0.01),the experimental group AST a significant decline in the M group(P < 0.01),E1 LDH is lower than the M group,but there is no significant difference(P > 0.05),E2 and E3 group was significantly lower than M group,each group CK and M group is significantly reduced,the E1 group significant differences compared with M group(P < 0.05),E2 and E3 group of extremely significant differences compared with M group(P < 0.01).(6)ELISA to detect immune cytokine levels: group M INF-gamma,serum TNF alpha in mice significantly decline in the C group(P < 0.01),compared with group M,IL-4 E1 group mice were significantly decreased(P < 0.05),the E1 group mice serum IL-10 cells express and M group was no significant difference(P > 0.05);E2 and E3 IL-4 groups of mice and IL-10 cells compared with M group significantly reduced(P < 0.05),serum E2 group mice INF gamma,TNF alpha significant rise in the M group(P < 0.01),and significantly lower than that of group C(P < 0.05);The INF-gamma in E3 group was significantly higher than that of M group(P < 0.01),and TNF-alpha was significantly higher than that of M group(P < 0.05),and the levels of inf-gamma and TNF-alpha in the E3 group were significantly lower than that in group C(P < 0.05).(7)Real-time PCR detection mRNA expression: d medicine ali red intervention,E2 mRNA and IL-4 M group comparison exists significant difference(P < 0.05),E1,E2 group decline trend in the M group,but there is no significant difference(P > 0.05);The E3 group of il-10 mrna was significantly lower than the M group(P < 0.05),and the E1 and E2 groups showed a decrease in group M,but there was no significant difference(P > 0.05).The comparison of inf-gamma,TNF-alpha mRNA and M group showed significant increase(P < 0.05).(8)Western blot detection,B7-2 protein expression B7-1: M group of mice spleen B7 1 protein expression is a significant increase in group C(P < 0.01),the trend of decline in the M group,while the E1 and E2 group compared with M group there were significant differences(P < 0.05),E3 group compared with M group is significantly decreased(P < 0.01).The expression of the spleen b7-1 protein in the spleen of mice was significantly decreased with the intervention of the vitamin.Conclusion:(1)Ali red water decoction can effectively promote the sports fatigue growth rate in mice,increase the thymus index and spleen index in mice and aerobic exercise ability,BLA,T,and at the same time reduce the exercise fatigue mice serum myocardial enzyme(AST,LDH and CK)level.(2)d medicine ali red water decoction can effectively correct the mouse movement state of immunosuppression,lower serum Th2 cytokines(IL-4,IL-10)level and the spleen IL 4,IL-10 mRNA expression level,and significantly higher serum Th1 cytokines(INF-gamma,TNF alpha)level and the spleen INF gamma,TNF alpha mRNA expression level,balance the body Th1 / Th2 immune balance.At the same time,the expression level of spleen B7-1 and B7-2 protein in the spleen of exercise fatigue mice was down-regulated.
