Effects On Metabolomics Of Maintaining Cardiac Function By Utilizing Acupuncture Pre-conditioning Therapy On Treating Exercise-induced Fatigue Rats

Objectives This study is based on the previous work on the effects of different acupoints on myocardial ischemia.Taking exercise-induced fatigue model rats as the research object,using the method of "acupuncture pretreatment",we selected the "health acupoint" group compatible with "Guanyuan" and "Zusanli" and "Guanyuan","Zusanli" and "Neiguan" acupoints compatible with "specimen acupoint" group and blank group and exercise group.The effects of CGRP and TERT on exercise-induced fatigue markers,myocardial endocrine function and antioxidant activity were compared.The potential biomarkers of CGRP and TERT were identified by 1H-NMR metabonomics.It provides new ideas and exploratory experience for the beneficial development of acupuncture and moxibustion in the field of sports medicine.Methods Sixty eight-week-old male SD rats,weighing 180-220 g,were randomly divided into four groups: blank group,exercise group,health care point group,and sample matching point group,with 15 rats in each group.Blank group: normal feeding,no exercise,no acupuncture;Exercise group: only exercise,no acupuncture;Health acupoint group: acupuncture pretreatment was conducted one hour before exercise every day,and the bilateral zusanli and guanyuan acupoints were selected.Specimens and acupoint matching group:acupuncture pretreatment was conducted one hour before exercise every day,and the neiguan,zusanli and guan yuan acupoints on both sides were taken.Method of acupuncture operation: bilateral "neiguan" point: the medial side of the rat forelimb,about 3mm between the ulnar and radial sutures saved from the wrist,and 2mm straight acupuncture;Bilateral "zusanli" point: the posterolateral of the knee joint of the rats,about 5mm below the fibula head,was stabbed for 3 ~ 5mm.White line "guan yuan" : rat’s abdomen,under the navel center about 2 mm,to xiphoid process direction of Angle of 45 ° inclined stab 3 mm;After routine skin disinfection,the skin was treated with 0.30 mm x 15 mm stainless steel filiform needle of huatuo brand,5mm straight acupuncture at each point,1min twisting at each point and 15 mim retention acupuncture for a total of 6weeks.In addition to the blank group,each group of six weeks of increasing load training,the last day at training,immediately after the training,the rat abdominal cavity anesthesia,apex department take blood extract serum,myocardial tissue,fully automatic biochemical instrument detection lactic acid(LD),creatine kinase(CK),urea nitrogen(BLU),malondialdehyde(MDA)and glutathione peroxidase(gsh-px)and superoxide disproportionation enzyme(T-SOD),ELISA method to detect the heart(ANP),sodium fluorescence quantitative PCR detection of myocardial mitochondrial oxidative damage,Western Blot analysis of calcitonin gene-related peptide(CGRP)and telomerase reverse transcriptase(TERT).1h-nmr was used to detect the metabolomics of the serum samples of rats in each group,and the data results were statistically analyzed.Results(1)compared with the blank group,the serum LD,CK and BUN contents in the exercise group were significantly increased(P<0.05).Compared with the exercise group,the contents of serum LD,CK and BUN were significantly decreased in the health point group and the sample matching point group(P<0.05).The decrease was more obvious in the point matching group than in the health point group(P<0.05).(2)compared with the control group,MDA content in the myocardial tissue of the exercise group was significantly increased(P<0.05),while t-sod and gsh-px content were significantly decreased(P<0.05).Compared with the exercise group,MDA content in myocardial tissue was significantly decreased in the health point group and the sample matching point group(P<0.05),and the content of t-sod and gsh-px was significantly increased(P<0.05).Compared with the healthy acupoint group,the content of MDA was significantly decreased(P<0.05),and the content of t-sod and gsh-px was significantly increased(P<0.05).(3)the expression of mitochondrial ND1 gene in cardiomyocytes was significantly higher in the exercise group than in the control group(P < 0.01).Compared with the exercise group,the mitochondrial ND1 gene expression decreased in the health point group and the sample matching point group(P < 0.05 or P < 0.01).Compared with the health point group,the expression of mitochondrial ND1 gene decreased in the sample matching point group(P < 0.05).(4)compared with the blank group,the ANP content in the myocardial tissue of the exercise group was significantly increased(P<0.05).Compared with the exercise group,the ANP content of myocardial tissue in the health point group and the sample matching point group was significantly lower(P<0.05).The ANP content of myocardial tissue in the sample matching point group was significantly lower than that in the health point group(P<0.05).(5)compared with the blank group,the expression of TERT protein in myocardium increased in the exercise group(P < 0.05),while the expression of CGRP protein decreased in the exercise group(P <0.05).Compared with the exercise group,the expression of myocardial CGRP increased in the health point group and the sample matching point group(P < 0.05),while the expression of TERT protein decreased in the health point group and the sample matching point group(P < 0.05).Compared with the health point group,the expression of TERT protein decreased in the sample matching point group,while the expression of CGRP protein increased(P < 0.05).(6)pls-da pattern recognition was significantly superior to PCA in metabolic pattern recognition and serum biomarkers after comparative analysis.The pls-da score graph of serum showed that the metabolic profiles of the exercise group were different from those of the other three groups,with a tendency of separation,and aggregation was observed between the serum samples of each group.Lactic acid,glucose,urea,alanine and acetic acid may be potential biomarkers based on the metabolite profile with VIP score greater than 1.Conclusions(1)acupuncture pretreatment can significantly improve the markers of sports fatigue,proving that acupuncture pretreatment has an obvious effect on fighting sports fatigue,and the matching of acupoints of specimens is better than the matching of health points.(2)acupuncture pretreatment has obvious antioxidant effect on exercise-induced fatigue myocardial tissue,can reduce damage to myocardial cells caused by exhaustive exercise,and has obvious regulatory effect on cardiac endocrine function,and the matching of acupoints of specimens is better than the matching of health points.(3)acupuncture pretreatment can play a protective role for the heart under exercise fatigue by regulating the expression and activity of CGRP and TERT in myocardial tissue.(4)pls-da pattern recognition was significantly superior to PCA in metabolic pattern recognition and serum biomarkers after
