Objective: There are differences in the expression levels of TNF-?,DKK-1 and TGF-?1 in fibrotic diseases,and we tried to investigate whether TNF-?,DKK-1,and TGF-?1 interact during the development of DD in order to reveal potential therapeutic targets and provide a theoretical basis for clinical treatment and prevention of recurrence.Methods:The study protocol and informed consent form of this study were approved by the Ethics Review Committee of the Third Affiliated Hospital of Inner Mongolia Medical University(Baogang Hospital,Inner Mongolia)and signed with the patients.The experimental tissue for this study was a pathological palmar aponeurosis specimen obtained from the palms of 20 patients with Dupuytren's contracture.For comparison,we also obtained fragments of the palmar aponeurosis from seven patients with carpal tunnel syndrome as a control group.The samples were divided into three portions immediately after sampling.Immunohistochemical staining,light microscopy,PCR detection and western blotting techniques were uniformly performed.Results :(1)TNF-? expression was up-regulated: TNF-? m RNA was highly expressed in the pathological tissues of DD patients compared with CT,P < 0.05,demonstrating that TNF-? expression was increased in DD patients;(2)Dkk-1expression was down-regulated: Dkk-1 m RNA was lowly expressed in the pathological tissues of DD patients compared with CT group,P < 0.05,demonstrating that Dkk-1expression was decreased in DD patients;(3)TGF-?1 expression was up-regulated:TGF-?1 m RNA was highly expressed in the pathological tissues of DD patients compared with CT group,P < 0.05,demonstrating that TGF-?1 expression was increased in DD patients;(4)Pearson correlation analysis suggested that TNF-?expression was positively correlated with TGF-?1 expression,TNF-? expression was negatively correlated with DKK-1 expression,and TGF-?1 expression was negatively correlated with DKK-1 expression.Conclusion:The experimental data showed that TNF-? and TGF-?1 were highly expressed and DKK-1 was lowly expressed in the pathological tissues of DD patients,and there was a link between the three at the expression level,and TNF-?,DKK-1 and TGF-?1 had a role in the pathogenesis of DD,and there was a link between the three.For the study of the relationship between the three and their related signaling pathways,it is promising to provide therapeutic targets and combination basis for the prevention and early treatment of Dupuytren's contracture.
