Antler plate,which is the disc-shaped ossification material retained on the horn handle of male sika deer(or red deer)after saw antler,falls spontaneously before giving birth to new antler in the following spring and is also known as "deer tray","deer flower plate" and "deer seat".It can be used to effectively treat sore,swelling and toxin,blood stasis pain,fatigue internal injury,low back pain and so on.It has strong anti-fatigue,anti-oxidation,blood-supplementing,and hypoglycemic activities,and has the potential of in-depth development.Thro?gh the study of mammals,it is found that proteins and peptides play an important role in life activities.The content of protein in the antler plate is as high as 40%,which is one of the main functional components of the antler plate,which contains a mixture of natural small molecular proteins and collagen,etc.,but the smaller the molecular weight of the protein is,the easier it is to be absorbed by human skin.Moreover,the skincare industry usually hydrolyzes collagen to obtain collagen peptides with better transdermal absorption and lower molecular weight,so,This paper focuses on natural small molecular peptides and enzymatically hydrolyzed collagen peptides,acts on Ha Ca T cells,discusses their skincare effects,and provides a basic theoretical basis for the in-depth development and utilization of antler plates.In this experiment,the total peptide desalination product of sika deer horn disk was prepared by acid extraction,alcohol precipitation,rotary evaporation,and gel desalination,and the small molecular protein mixture was prepared by reverse high-performance liquid chromatography(RP-HPLC).The mixture of small molecular peptides was analyzed by SDS-PAGE electrophoresis and mass spectrometry.On the other hand,collagen was extracted from the antler plate of sika deer by the hot water method,and the collagen in the antler plate was hydrolyzed by trypsin with the degree of hydrolysis of collagen as an index.The preparation process of collagen peptide was optimized by a single factor test and the Box-Behnken design test.The collagen peptide was analyzed by SDS-PAGE electrophoresis and mass spectrometry to determine its yield and relative molecular weight distribution.On this basis,the mixture of small molecular polypeptides,collagen peptides and collagen from antler plate were treated on Ha Ca T cells,and their effects on the morphological changes of Ha Ca T cells,the proliferation of Ha Ca T cells and the changes of Superoxide dismutase(SOD)activity,hydroxyproline(Hyp)content and Malondialdehyde(MDA)content of Ha Ca T cells were studied to explore their skincare ability.The results showed that the molecular weight of the small molecular polypeptide mixture of sika deer horn disk was about 10 k Da detected by SDS-PAGE electrophoresis and mass spectrometry,and the relative molecular weight of collagen peptide prepared by trypsin hydrolysis was mainly below 5k Da by electrophoresis and mass spectrometry.The collagen content of the sika deer horn disc was 22.46%.The optimum preparation process of deer antler collagen peptide was enzymatic hydrolysis time 2.2h,p H 8.0,temperature 50?,enzyme amount 1200 U/g.Under this condition,the degree of protein hydrolysis was 15.97%.The effects of collagen protein of antler plate,the mixture of small molecular polypeptides of antler plate and collagen peptide of antler plate on skincare were compared.it was found that all of them could affect Ha Ca T cell morphology,cell proliferation,Superoxide dismutase(SOD)activity,hydroxyproline(Hyp)content and Malondialdehyde(MDA)content in different degrees,and at a concentration of 100?g / m L,the small molecule peptide action group has better performance than the collagen peptide action group in terms of improving SOD activity and promoting Hyp production,or inhibiting MDA production ability,and both are superior to collagen Proteome.This shows that the skin care effect of small molecule peptides is better.Therefore,the skin care effects of the three are determined from large to small: small molecule peptide> collagen peptide> collagen.
