Acanthopanax senticosu(Rupr.et Maxim.)Harms(AH)is a perennial deciduous shrub of the genus Acanthopanax of the Araliaceae.Its dried roots and rhizomes or stems are included in the Chinese Pharmacopoeia as a traditional Chinese medicine.It has the effect of invigorating qi and spleen,nourishing the kidney and calming the nerves;its leaf is used as a local Chinese medicinal material and the standard is included in the 2001 edition of "Heilongjiang Provincial Standard of Chinese Medicinal Materials".Both leaves and fruits are raw materials for processing Acanthopanax senticosus tea,and they often drink to calm down and help sleep.The Acanthopanax sessiliflorus(Rupr.et Maxim.)Seem(AS),which has the same family and the same genus,has a similar appearance to AH,and there is a certain mixed phenomenon.For example,the root bark of the two is used as the source of the local medicinal material,And Acanthopanax senticosus tea is often mixed.In recent decades,the quality of Acanthopanax senticosus medicinal resources has declined significantly due to the unorganized digging of stems and roots,and even the phenomenon that no qualified medicinal materials are available.In this paper,we used Acanthopanax senticosus leaves(AHL),Acanthopanax senticosus fruits(AHF),Acanthopanax sessiliflorus leaves(ASL),and Acanthopanax sessiliflorus fruits(ASF)as experimental objects,respectively.The water extract(WE)and ethanol extract(AE)of the above raw materials were carried out in terms of sedative hypnotic activity and mechanism a comparative analysis is provided to provide a basis for the sustainable development and utilization of AH and AS resources.1.Study on the sedative and hypnotic activity and mechanism of water and alcohol extracts of AHL and AHF.Prepare AHL water extract(AHL-WE)and alcohol extract(AHL-AE)and AHF water extract(AHF-WE)and alcohol extract(AHF-AE)respectively.By observing the effects of AHL-WE?AHL-AE?AHF-WE and AHF-AE on the autonomous activities of mice,and combined with the subliminal hypnotic dose of pentobarbital sodium(28 mg/kg,i.p.)and the hypnotic dose(48 mg/kg,i.p.)on the sleep of mice,the sedative and hypnotic effects of AHL-WE?AHL-AE?AHF-WE and AHF-AE were investigated,and the sedative and hypnotic dose and subhypnotic dose of AHL-WE?AHL-AE?AHF-WE and AHF-AE were determined.To investigate the sedative-hypnotic mechanism of AHL-WE?AHL-AE?AHF-WE and AHF-AE through the combined administration of AHL-WE?AHL-AE?AHF-WE and AHF-AE with 5-Hydroxytryptophan(5-HTP)and Flumazenil(FLU)and the effects of AHL-WE?AHL-AE?AHF-WE and AHF-AE on sleep and neurotransmitters in hippocampus of insomniac mice induced by p-Chlorophenylalanine(pCPA).Result: Among AHL-WE?AHL-AE?AHF-WE and AHF-AE could reduce the number of spontaneous activities in mice,increase the sleeping rate of mice with subthreshold dose of sodium pentobarbital,shorten the latency of sleep and prolong the duration of sleep in mice with synergistic hypnotic dose of sodium pentobarbital,showing sedative and hypnotic effects.The hypnotic dose and sub-hypnotic dose were 8 mg/kg and 32 mg/kg,respectively.Subhypnotic dose of AHL-WE?AHL-AE?AHF-WE and AHF-AE combined with subeffective dose of 5-HTP(2.5 mg/kg,i.p.)could significantly reduce sleep latency and prolong sleep time in mice,and significantly increase 5-Hydroxytryptamine(5-HT)level in hippocampus of mice.pCPA(100 mg/kg,i.p.)could significantly inhibit the sedative and hypnotic effects of AHL-WE?AHL-AE?AHF-WE and AHF-AE at hypnotic doses and the elevation of 5-HT in hippocampus of mice.FLU(8 mg/kg,i.p.)and TSC(8 mg/kg,i.p.)effectively antagonized the sedative and hypnotic effects of AHL-WE?AHL-AE?AHF-WE and AHF-AE and the increase of g-Aminobutyric acid(GABA)level in hippocampus of mice.The results show that AHL-WE,AHL-AE,AHF-WE and AHF-AE have better sedative and hypnotic effects,of which AHF-AE is the strongest sedative and hypnotic activity,sedative and hypnotic activity of AHF is more stable than AHL and each the sedative and hypnotic effects of alcohol extracts in medicinal sites are better than water extracts,and its mechanism of action may be related to the central nervous system of 5-HT and GABAergic.2.Study on the sedative and hypnotic activity and mechanism of water and alcohol extracts of ASL and ASF.Prepare ASL water extract(ASL-WE)and alcohol extract(ASL-AE)and ASF water extract(ASF-WE)and alcohol extract(ASF-AE)respectively.Using the same experimental methods and experimental conditions as in the previous chapter,the sedative and hypnotic activities of ASL-WE,ASL-AE,ASF-WE,and ASF-AE were investigated.The results show that the sedative hypnotic effect of AS extract is not as significant as that of AH extract,but it also has good sedative hypnotic activity.In general,the order of sedative hypnotic activity is ASF-AE>ASF-WE>ASL-AE>ASL-WE,and may have a hypnotic effect by affecting the 5-HT and GABAergic central nervous system.
