In this article, method of pharmacognosy was applied to carry out the macroscopic,microscopic, physical and chemical identification of acanthopanax senticocous leaves;Ultraviolet spectrophotometry was used to determined the content of flavonoids inacanthopanax senticocous leaves; High-performance liquid chromatography (HPLC) wasadopted to the content of nucleoside st. John’s wort in acanthopanax senticocous leaves.Results showed that the content of flavonoids and st. John’s wort glucoside was0.06%and0.029%, respectively. According to the one part of Chinese pharmacopoeia2010version,content of water and ash was determined in the leaves. The established method for thedetermination was of easy operation, high specificity and could be used as the quality controlstandard of acanthopanax senticocous leaf, also could provide basis for the development of it.The spray drying was applied to dry acanthopanax senticocous leaves, single factorscreening tests was carried out first, then, material liquid density, inlet wind temperature,compressed air flow and the speed of peristaltic pump were selected as the spray dryingprocess factors, yield and water content of powder were selected as the indexes, the optimaltechnological conditions of orthogonal experiment was as follows: the relative density of thesolution was1.20, inlet wind temperature was190℃, compressed air flow was600L/h, thespeed of peristaltic pump was20%. The powder was of nice color and with little water.The orthogonal experimental was used to optimize the dropping pill molding process. Onthe study of the prescription ratio and the dropped temperature, the difference of pill heavy,dissolve scattered time limit, sensory evaluation (the same length pellets, hardness, colour andlustre) were selected as the indexes. The optimal process was A2B2C3, namely, the ration ofdrugs to matrix was1:3, Matrix A to matrix B was1:2, the dropped temperature was95℃.On the study of mixture ratio of simethicone and liquid paraffin wax, temperature and thecondensing column height, weight difference, dissolved in pills scattered time limit, theappearance quality (the same length pellets, hardness, trailing, colour and lustre) wereselected as the indexes. The optimal process was A3B2C2, namely, the ratio of simethiconeand liquid paraffin wax and liquid paraffin wax was1:3, temperature of cooling agent was4℃, cooling column height was60cm.The weight difference, scattered time limit, stability, TLC identification of the main ingredient nucleoside st. John’s wort of the acanthopanaxsenticocous leaves dropping pill were investigated. And the content determined method formain components was established. The results showed this product was brown with smoothsurface, rounded shape color uniformity at normal temperature. Weight difference of all threebatches of samples was less than3%; all scattered time limit was within30min; Theflavonoids content was set as no less than115mg/g; St. John’s wort content was set as no lessthan9mg/g. Results of stability test showed that the pill was stable at room temperature (25℃±2℃) as well as after accelerated (40±2℃,75+5%) placed for6months, theappearance is brown, complete, the color and luster is even, hardness was suitable; weightdifference and scattered time limit didn’t change obviously; The content flavonoids and st.John’s wort glucoside was stable.Study on pharmacodynamics showed that the flavonoids in acanthopanax senticocousleaves could significantly reduce the neurological symptoms of brain arteryischemia-reperfusion model rats, reduce thecerebral water content and brain index, reducewhole blood viscosity and plasma viscosity.There was no obvious difference between it andthe positive control medicine step length NaoXinTong capsule about the above action, whichindicated that the flavonoids in acanthopanax senticocous leaves could protect the rat withMCA ischemia-reperfusion injury. The experimental result of acute toxicity showed thebiggest tolerance was12g/kg and there was no obvious toxic reaction.
