Objective:In this study,we observed the improvement of endometrial receptivity in f emale rats with PCOS model by tonifying kidney and resolving phlegm throug h pinopodes process and endometrial regulatory factors.Methods:A total of 123 female rats with SPF grade SD were selected at the age o f 7 to 8 weeks.Vaginal smear was used to screen 120 female rats with norma l estrus cycle.Twenty rats were randomly reserved as the blank control group,and the remaining rats were given letrozole combined with high-fat emulsion for 28 days to establish the obese PCOS animal model.The successful model rats were selected and randomly average divided into model control group,low,medium,high and metformin dose group?The low,medium and high dose groups were given the same volume of t he corresponding concentration of traditional Chinese medicine,the metformin group was given equal volume metformin suspension,and the blank group was given the same volume of distilled water.Weight was calculated after 21 da ys of medical treatement,the ovarian tissue and uterine tissues were taken out for HE staining,observation of Pectinate under Electron Microscopy,the serum level of E2,PROG and T were detected by Elisa method,the expressions of P R,integrin?v?3 and E2R were detected by immunohistochemical staining m ethod,The m RNA expression of Cyclin D1 was detected by q PCR method.The data was processed by SPSS25.0 statistical software.Results:1. Effect of KNPR on the estrous cycle and body weight of the obese PC OS rats:Compared with the blank control group,the estrous cycle of the fema le rats in the model control group was disordered and their body weight was i ncreased significantly?P<0.01?.Compared with the model control group,the estrous cycle of the high-dose Chinese medicine group was compared with that of the low-dose Chinese medicine group.Regularity,the body weight of each group of Chinese medicine decreased significantly?P<0.01?.2. Effect of KNPR on serum sex hormones of obese PCOS rats:Compare d with the blank control group,the serum PROG and E2concentration in mod el control female rats were decreased?P<0.05 or P<0.01?and the serum T concentration was increased?P<0.01?;Compared with the model control grou p,the serum PROG and E2concentration in the high-dose group of Chinese medicine were increased?P<0.05?,and the serum T concentration was decreas ed?P<0.05?.There was no statistical significance in the serum PROG,T,an d E2concentration of the low-medium group of Chinese medicine.3. Effect of KNPR on the development of ovaries and follicles of the obe se PCOS rats:compared with the blank control group,the interstitial fibrous ti ssue in female ovaries proliferated,the follicular granulosa became thinner,the follicular membrane cell layer significantly thickened,the number of atretic fo llicles increased,and the number of corpus luteum decreased in the model cont rol group;Compared with the model control group,the follicular granulosa cell layer was thickened and the number of atretic follicles was reduced.Primary,secondary and even mature follicles at different developmental stages could be seen.The number of corpus luteum was increased in the high dose group.4. Effect of KNPR on uterine morphology and development of eosinophil s in obese PCOS female rats:Compared with the blank control group,the eosi nophilic process in the model control group is in the developing stage,the eos inophilic process is in the developing stage,and the uterus is in the pre-estrus stage.A large number of endometrial epithelial cells degeneration and necrosis,punctate necrosis of lymphocytes and glandular epithelial cells,nuclear shrinka ge,and the number of glands was decreased significantly?P<0.01?.Compared with the model control group,the cytospora of the high-dose group of Chines e medicine is in the mature stage,and its morphology is similar to that of the blank group.The cytospora of the middle-dose group of Chinese medicine is at the late stage of development.There are a large number of neurites in the early stage of development;the positive group is in the esophageal process an d is in the late stage of development.There are a small number of mature eso phageal processes and a large number of developing esophageal processes.The endometrium of the high-dose group is in estrus.Edema,low-medium dose of Chinese medicine is in the pre-estrus stage,lymphocyte infiltration,lamina pr opria edema;positive group is in the pre-estrus stage,lymphocytes and neutrop hils infiltrate endometrial epithelial cells degeneration and necrosis.The number of glands in each group of Chinese medicine was increased significantly?P<0.01?.5. Effect of KNPR on uterine receptivity-related molecules in obese PCOS female rats:Compared with the blank control group,the expressions of integri n?v?3 and Cyclin D1 m RNA in uterine receptivity-related molecules of the model control group was decreased?P<0.05?,The expressions of PR and E2R were increased?P<0.05?.Compared with the model control group,the express ion of integrin?v?3 and Cyclin D1 m RNA in the high-dose group of Chines e medicine were increased?P<0.05 or P<0.01?.The expressions of PR and E2Rwas decreased?P<0.01 or P<0.05?.The integrin?v?3,Cyclin D1 m RN A,PR,and E2R in the low-dose group of Chinese medicine had no statistical s ignificance.Conclusion:1.The method of KNPR can regularize the estrous cycle of females with obese PCOS model,control the continuous weight gain of rats,and thereby im prove the obesity of female PCOS rats.2.The method of KNPR can correct the reproductive endocrine disorders of female rats with obese PCOS model,improve the state of Kaohsiung,and promote the development of the ovary.Establish a good endometrial environme nt through the pectin,E2R,PR,and integrin?v?3;The effect of improving endometrial receptivity is exerted.3.The method of KNPR may play a mechanism of improving endometri al receptivity by regulating sex hormones and activating Cyclin D1-related signal ing pathways.
