[Objective]To investigate the effect and mechanism of adiponectin(APN)on endometrial receptivity(ER)in polycystic ovary syndrome(PCOS)rats.[Method]First of all,female SD rats were randomly divided into control group(CON group),normal saline group(NS group)and letrozole group(LET group).In the LET group rats,letrozole was successfully used for intragastric administration to establish PCOS rats model.Then,PCOS rats were treated by recombinant adiponectin(APN),recombinant adiponectin+GW6471(PPAR?specific inhibitor),or recombinant adiponectin+corn oil,respectively.Subsequently,we have detected changes in the estrous cycle,average body weight,ovarian morphology,ovarian index,uterine morphology,uterine index,levels of serum hormones and levels of lipid metabolism.Moreover,Immunohistochemistry and Western blot were used to detect the expression of PPAR?and HOXA10 of the endometrium in each group of rats.Finally,we have observed the development of endometrial pinopode(PP)in the estrus of each group by scanning electron microscopy and examined fertility,by female rats mating with proven stud males.[Result]1)Compared with the control(CON)group rats,the letrozole(LET)group rats had estrous cycle disorders,higher average weight,polycystic ovarian changes,increased ovarian index,slender uterine,decreased uterine index,serum hormones disorders and serum lipids disorders.Moreover,no significant changes in the normal saline(NS)group.These have suggested that the PCOS rats model is successfully established.2)Compared with the CON group rats,the endometrial PPAR?and HOXA10 expressions in the PCOS rats were significantly reduced(P<0.05),while the NS group rats had no significant changes in the endometrial PPAR?and HOXA10 expressions(P>0.05).3)Compared with the CON group rats,the experimental group1(PCOS+APN group),the experimental group 2(PCOS+APN+GW group)and the positive control group(PCOS+APN+VEH group)rats gradually recovered to normal estrous cycle and ovaries.While the estrous cycle of PCOS group rats,the negative control group,was disordered,and the ovaries of these were still polycystic.The average weight and ovarian index of the PCOS group rats were significantly higher than the CON group rats,and the average uterine index was significantly lower than the CON group rats.However,compared with the PCOS group rats,the body weight and ovarian index of the PCOS+APN group rats were significantly lower,and the rat uterine index was significantly higher(P<0.05).Compared with the PCOS group rats,the serum hormone and lipid metabolism of the PCOS+APN group rats has improved.4)Compared with the CON group rats,the expressions of PPAR?and HOXA10 in the endometrium of the PCOS group rats were significantly reduced(P<0.05).Compared with the PCOS group rats,the expressions of PPAR?and HOXA10 in PCOS+APN group rats were significantly increased(P<0.05).Compared with the PCOS+APN group rats,the expressions of PPAR?and HOXA10 in the PCOS+APN+GW group rats were significantly decreased(P<0.05).Compared with the PCOS+APN group rats,there were no statistical differences in the expressions of PPAR?and HOXA10 in the PCOS+APN+VEH group rats.Compared with the PCOS+APN+GW group rats,the expressions of _IP_VPAR?and HOXA10 in the PCOS+APN+VEH group rats were significantly up-regulated(P<0.05).5)Compared with the CON group rats,the endometrial PP of the PCOS group rats was poorly developed.Compared with the PCOS group rats,the PP of the PCOS+APN group rats developed well.Compared with the PCOS+APN group rats,the PP of the PCOS+APN+GW group rats was poorly developed.Compared with the PCOS+APN+GW group rats,the PP of the PCOS+APN+VEH group developed well.6)Compared with the PCOS group rats,the pregnancy rate of the rats in each group was the same.Though,the number of embryos was significantly reduced and the development was retarded in the PCOS+APN+GW group rats,compared with the PCOS+APN group rats.[Conclusion]Adiponectin(APN)can improve endometrial receptivity(ER)in PCOS rats through the PPAR?/HOXA10signaling pathway.Adiponectin or PPAR?could be alternative strategies for ameliorating pregnancy outcomes in patients with PCOS.
