Objectives To investigate the effect of visfatin on the endometrial receptivity of SDrats during implantation. By detecting the expression level of VEGF and eNOS on theendometrium during the implatation period, to further explore the role of visfatin onendometrial receptivity and find it possible mechanism.Methods According to aging,120female SD rats were divided into two groups: Sixtyolder rats(32~36weeks) were included in Group1and sixty younger rats(12~16weeks)were included in two Group, all the sixty mice in each group were randomized into thetreatment group (surperstimilation+visfatin) and the control group(surperstimilation),30cases in every group. After successful modeling,calculate the pregnancy rate andmean implanted blastocysts. observed the endometrial histology with OpticalMicroscope. Detection of in endometrial with Polymerase Chain Reaction and Real-time Polymerase Chain Reaction.HE staining was used to survey the endometriummorphology.At the same time, immunohistochemistry (IHC) was used to test themicrovascular density of endometrium. Application of rt-pcr to detect the expression ofVEGF and eNOS. Experimental data were statistically analyzed using SPSS17.0software.measurement data as mean±standard deviation (x s) that the use of single-factoranalysis of variance to determine group differences between the two groups werecompared using S-N-K test data. P <0.05was considered statistically significant. VEGF,eNOS correlation analysis using linear correlation.Results1The results of electron microscope indicate the endometrial epithelial cellsconnect closely, pinocytosis fully developed, quantity, development is synchronous,morphological rules, the surface is smooth, outburst pinocytosis vesicles is rich,organelles and glycogen granules in young intervention group and non-intervention grouptwo groups; Older intervention group compared with the intervention group, endometrialepithelial cells connect closely, pinocytosis fully developed, quantity, development issynchronous, morphological rules, the surface is smooth, outburst pinocytosis vesicles isrich, organelles and glycogen granules.2Immunohistochemical results in interventiongroup and intervention group two groups of young endometrial capillaries density, therewas no statistically significant difference (P>0.05); Older intervention group of endometrial capillaries density higher than older than intervention group, the differencewas statistically significant (P <0.05).3young intervention group and the interventiongroup two groups of endometrial VEGF mRNA expression, there was no statisticallysignificant difference (P>0.05);Older intervention group endometrial VEGF mRNAexpression is higher than the intervention group, the difference was statisticallysignificant (P <0.05).4young intervention group and the intervention group two groupsof endometrium eNOS mRNA expression, there was no statistically significant difference(P>0.05);Older intervention group endometrial eNOS mRNA expression is higher thanthe intervention group, the difference was statistically significant (P <0.05).5VEGF andThe expression of eNOS mRNA in endometrial was positively correlated.6youngintervention group and the intervention group in the two groups of pregnancy rate, theaverage number of blastocyst, there was no statistically significant difference (P>0.05);Older pregnancy rate of the intervention group, the average number of blastocyst,higher than that of the intervention group, the difference was statistically significant (P <0.05).Conclusions1) the receptive age affect the lining of the uterus.2) within the exogenousfatty element can improve stimulate ovulation older rats "window period" farm inreceptive endometrium, but to improve the effect of the young rats and the receptivity ofendometrium was not significant.3) within the exogenous fat element can improvestimulate ovulation older mice pregnancy rate, the average number of blastocyst, but thepregnancy rate in the improvement of young rats, the average number of blastocyst, theeffect is not significant.4) within the exogenous fatty element may be through raising theexpression of VEGF and eNOS promote endometrial angiogenesis older SD rats, andthen improve the older mice and the receptivity of endometrium.
