Objective : Serum oxidative markers SOD,GSH,MDA and 8-Ohd G were detected in constipation patients,and compared with healthy population to analyze the oxidative stress reaction of constipation patients,and to preliminarily clarify the correlation between constipation and oxidative stress reaction and the relevant mechanism of constipation.Then,through the intervention of xuanshen decoction in patients with constipation,the relevant oxidative stress indicators were detected,the mechanism of xuanshen decoction was analyzed from the perspective of oxidative stress,and the clinical efficacy and safety evaluation of xuanshen decoction were observed,laying a foundation for subsequent related studies.Methods : 1.According to the inclusion criteria and exclusion criteria,the patients with chronic constipation and the healthy people will be enrolled.The plasma of the subjects and the centrifugal serum were used to detect the contents of SOD,MOD,GSH and 8-Ohd G with ELISA.By comparing the data differences between the experimental group and the control group judge whether there is any correlation between constipation and oxidation stress.2.After confirming the correlation between constipation and oxidative stress reaction,the second part of the experiment were started.According to the inclusion criteria and exclusion criteria,the patients with chronic constipation and healthy people were enrolled into the experiment group(constipation-Xuanshen-Tang group),the control group A(constipation-Procapride succinate group),the control group B(constipation-placebo group),and the blank group(healthy people group)took the subjects' plasma before being formally enrolled into the group The contents of SOD,mod,GSH and 8-OHd G were detected.After 30 days of intervention,the plasma of te subjects was taken again to detect the correlation,and the differences between four groups before and after intervention were compared,so as to determine the oxidative stress mechanism of Xuanshen-Tang.3.Patients with chronic constipation were divided into three groups: the treatment group(constipation-Xuanshen-tang group),the control group(constipation pulucapride succinate group)and the placebo group(constipation-placebo group).According to the Wexner constipation scoring standard,the scores of related symptoms,adverse reactions and acceptances were recorded before treatment and 7,15,30 and 15 days after treatment respectively,and the differences between the three groups were compared,To evaluate the clinical efficacy and safety of Xuanshen decoction.Results:1.The values of oxidation markers between healthy population and constipation patients were compared by t test,SOD(T=2.325,P=0.028>0.05),GSH(T=0.024,P=0.981<0.05)MDA(T=4.135,P=0.000<0.05),8-Ohd G(T=11.357,P=0.000<0.05).The difference of SOD value between healthy people and constipation group was statistically significant,while the difference of GSH value was not statistically significant,while the difference of MDA and 8-ohdg was statistically significant.Compared with healthy people,the serum antioxidant substances of constipation people are lower than that of healthy people,and the serum oxidative stress damaging substances are significantly higher than that of healthy people,so there is a clear correlation between the occurrence of constipation and oxidative stress response.2.In the second part of the experiment group,Welch-ANOVA test showed that SOD(F=1.534,P= 0.228)GSH(F=0.795,P=0.458),MDA(F=0.587,P=0.561),8-ohdg(F=0.694,P=0.505)were not significantly different in the constipation group before intervention,and there was no significant difference in the difference of P>.SOD value(F=10.038,P=0.000,P <0.05)GSH value(F=11.239,P=0.000,P <0.05),MDA value(F=1.512,P=0.000,P <0.05),8-ohdg(F=117.544,P=0.000,P <0.05)showed significant differences in oxidative stress indicators in the constipation group after intervention(P<0.05).Compared with the experimental group,the expression of GSH in control group A was lower than that in the experimental group,and theexpression of MDA and 8-ohdg was higher than that in the experimental group,the difference was statistically significant(P<0.05),while the expression of SOD and GSH in control group B was lower than that in the experimental group,and the expression of 8-ohdg was higher than that in the experimental group,the difference was statistically significant(P<0.05).Compared with control group A,the expression level of MDA and 8-ohdg in control group B was also lower than that in control group A,with statistically significant difference(P<0.05).The antioxidant expression of the experimental group was higher than that of the control group A and B,and the oxidative damage was lower than that of the control group A and B.Compared with placebo,the level of oxidative damage in control group A was also higher than that in placebo,which may aggravate oxidative damage and constipation.(4)experimental group compared before and after the intervention,SOD value(T = 2.509,P = 0.025)GSH value(T = 3.779,P = 0.002),MDA value(T = 2.713,P = 0.017),8-Ohd G value(T = 18.191,P = 0.000,P < 0.01)after intervention figwort root soup,oxidative stress indicators in statistically significant difference(P < 0.05)figwort decoction can improve the body's antioxidant factor expression of SOD and GSH in serum,reduce oxidative damage factor MDA,significantly reduced the expression of 8-Ohd G.(5)SOD value(T=0.740,P = 0.471)GSH value(T=2.015,P =0.063,P >0.05),MDA value(T=0.469,P=0.646,P >0.05),8-ohdg(T=2.432,P =0.029,P <0.05)after succinic acid propidium prubicide intervention,the changes of SOD,GSH,MDA markers had no statistical significance(P >0.05),but it increased the expression of oxidative damage factor 8-ohdg(P<0.05).3.Compared with the control group,the treatment effect of the two groups was similar,and the score was similar,both of which were better than the placebo group(P<0.05).In the comparison of symptoms,the treatment group and the control group were effective in improving fecal properties,slowing down defecation effort,and reducing defecation abdominal distension,which were positive to the treatment time.The three groups had no significant effect on reducing the time of single defecation.The control group was better than the treatment group and the control group in improving the frequency of defecation.In terms of drug acceptance,both the treatment group and the placebo group were significantly higher than the control group(?2=11.100,P=0.004).Conclusions:1.Constipation is caused by internal and external factors,which reduces the body's antioxidant capacity and increases the oxidative stress response,leading to the body's cell tissue damage.2.Xuanshen-Tang can enhance the anti-oxidation ability of the body in patients with constipation and reduce the oxidative stress injury,which may be related to the improvement of SOD and GSH expression,the decrease of MDA and 8-ohdg expression,so as to restore the balance of oxidative stress.3.Xuanshen-Tang has a definite clinical effect,which can effectively improve the fecal characters of patients with functional constipation,relieve the effort of defecation,and relieve the degree of abdominal distension.4 procarpiril succinic acid can increase the expression of 8-ohdg in serum of patients with chronic constipation to a certain extent.Long-term use may increase oxidative stress injury and increase the risks of constipation.
