Objective:To explore the effects of HIF-1?,analyze the urinary 8-OHdG to reflect the degree of oxidative stress injury,and evaluate the predictive value of serum HIF-1? and urinary 8-OHdG for early pregnancy loss.Method:The early pregnant who underwent artificial abortion at Qingdao Municipal Hospital from November 2018 to June 2019 were collected and recorded.42 cases of early pregnancy loss who met the criteria were included as the study group.And 48 cases of normal early pregnancy requiring induced abortion during the same period were randomly selected as the control group.The serum HIF-1? and urinary 8-OHdG were detected with ELISA,and the 8-OHdG was corrected by the detected creatinine level.The HIF-1? protein was detected with Western-Blot,while the HIF-1? mRNA with RT-PCR.Using statistical software to analyze the experimental results and draw a conclusion,and P <0.05 was considered statistically significant.Result:1.Basic data: There was no significant difference in age and gestational weeks between the study and control group(P>0.05).2.Changes of HIF-1? levels between two groups: In the study group,the serum HIF-1? level was significantly lower than that of the control group(5573.01 793.43 pg / ml <6488.95 831.09 pg / ml),the difference was statistically significant(P <0.001);the expression of HIF-1? protein in the villous tissue was significantly lower than that of the control group(1.28 0.31 <1.51 0.45,P <0.01).Compared with the control group,the HIF-1? mRNA of the villous tissue in the study group showed decreased,but the difference was not statistically significant(0.77 0.39 <1.35 1.18,P > 0.05).3.Changes in urinary 8-OHdG levels between two groups: The urinary 8-OHdG in the study group was higher than the control group(18.15 3.44ng/mgCr> 14.68 3.97 ng / mgCr),and there was a statistically significant difference(P <0.001).4.Correlation analysis of serum HIF-1? and urinary 8-OHdG levels: The serum HIF-1? and urinary 8-OHdG were lowly negative correlated(r=-0.346,P<0.01).5.Analysis of the predictive value of serum HIF-1?,urinary 8-OHdG and the combination of both for early pregnancy loss: In the ROC curve,the sensitivity of serum HIF-1?: urinary 8-OHdG: the combined index was 83.3%: 60.42%: 85.4%;specificity was 64.3%: 83.3%: 69.0%.The AUC analysis was 0.816: 0.751: 0.837.Conclusion:1.Maternal oxidative stress injury can cause early pregnancy loss.2.In the local villous tissue of the early pregnancy loss,the time-dependent oxidative stress response induced by HIF-1? was abnormal.3.Maternal HIF-1? can negatively regulate urinary 8-OHdG level to aggravate the oxidative stress response,and urinary 8-OHdG level can further reflect the degree of oxidative stress damage of the early pregnancy loss.4.For the evaluation of oxidative stress injury in early pregnancy loss,the relative sensitivity of serum HIF-1? combined with urinary 8-OHdG is higher,which can guide the early clinical prediction,diagnosis and treatment.
