Experimental Study On The Effect Of A-PRF On The Survival Rate Of Rabbit Fat Transplantation

Abstract/Summary:
Objective: To explore the application of advanced platelet-rich fibrin(A-PRF)and common platelet-rich fibrin(PRF)in fat transplantation in rabbit.Provide guidance for clinical applications.METHODS: Eighteen New Zealand experimental rabbits were randomly divided into three groups 。 The adipose tissue on the back of the rabbits was removed,and the surface fascia was removed and cut into pieces for use.Blood was collected from rabbit ear middle arteries,and common PRF was prepared by centrifugation at 3000 r / min for10 min,and A-PRF was prepared by centrifugation at 2000 r / min for 24 min.The fat was mixed with different types of PRF and transplanted into the area of rabbit ear exfoliation.Experimental group(A-PRF group): 2ml A-PRF + 1ml granular fat;positive control group(PRF group): 2ml PRF + 1ml granular fat;negative control group(NS group): 2ml saline + 1ml granular fat.At 4 weeks,8 weeks,and 12 weeks after transplantation,samples were taken for gross observation,specimen weighing,HE staining,and immunostaining to compare fat survival rates and neovascularization differences among the three groups.RESULTS: The weight difference of each group was statistically significant at different time points(P <0.05).The fat specimens in the A-PRF group and the PRF group were heavier than the NS group,and the fat survival rate in the A-PRF group was significantly higher than that in the PRF group.HE staining showed that the size and distribution of adipocytes in the A-PRF group were more uniform at different time points,and there were more infiltration of inflammatory cells between the groups.The number of inflammatory cells infiltration in the A-PRF group was higher than that in the other two groups.Immunohistochemistry showed that the number of new capillaries in the A-PRF group was significantly better than that in the two control groups,and the results in the PRF group were better than those in the NS group.Conclusion: A-PRF is superior to common PRF in increasing the survival rate of adipocytes,reducing inflammation and promoting capillary angiogenesis,which provides a reference for the use of A-PRF in clinic.
