ObjectiveTo investigate the expression of Irisin in endometrioid endometrial carcinoma and its effect on endometrial carcinoma Ishikawa cells.MethodsWestern blot and real-time quantitative PCR were used to detect the expression of irisin in 36 cases of endometrioid endometrial carcinoma and 36 cases of normal endometrium in Qingdao municipal hospital affiliated to Qingdao university from August 2016 to March 2018.The effects of irisin on the migration and invasion ability of Ishikawa cells were detected by scratch experiment and Transwell chamber invasion experiment.The effect of irisin on the expression of e-cadherin,Vimentin and Snail in Ishikawa cells was detected by Western blot.Results1.The expression levels of irisin protein and mRNA in endometrioid endometrial carcinoma tissues were higher than those in normal endometrium tissues(P < 0.001,P < 0.001).The expression level of irisin protein in endometrioid endometrial carcinoma was correlated with the depth of infiltrating muscle layer(P=0.004),lymph node metastasis(P < 0.001),and pathological grade(P=0.017),but not with age(P=0.130).The expression level of irisin mRNA was related to the depth of infiltrating muscle layer(P < 0.001)and lymph node metastasis(P=0.002),but was not related to pathological grade(P=0.086)or age(P=0.238).2.The scratch test results showed that the healing rates of irisin 25nmol/L,50nmol/L and 100nmol/L groups were 45.40±2.91,61.93±3.43 and 66.70±4.76 respectively,higher than that of the control group(26.80±1.18).The difference was statistically significant and showed a concentration dependence(P < 0.001).Transwell invasion assay showed that the cell Numbers of irisin 25nmol/L,50nmol/L and 100nmol/L groups were 227.00±52.03,295.33±26.10 and 348.00±35.38 respectively,which was significantly higher than that of the control group(92.33±20.26),and the difference was statistically significant and concentration-dependent(P < 0.001).3.Western blot experiment showed that the expressions of Snail and Vimentin in irisin groups 25nmol/L,50nmol/L and 100nmol/L were 1.21±0.11,1.20±0.12,1.39±0.20,1.31±0.12,1.70±0.19,1.46±0.10 respectively,which were higher than those in the control group(0.90±0.08,1.01±0.05,P=0.001,P=0.004).The expression levels of e-cadherin in irisin groups 25nmol/L,50nmol/L and 100nmol/L were 0.88±0.18,0.68±0.13 and 0.48±0.09 respectively,which were lower than those in the control group(1.23±0.11),and the difference was statistically significant(P=0.001).Conclusion1.The expression level of irisin in endometrioid adenocarcinoma tissues was significantly higher than that in normal endometrial tissues,and the expression levels of irisin protein and mRNA were related to the depth of infiltrating muscle layer and lymph node metastasis.2.Irisin can can promote the migration and invasion ability of Ishikawa cells by inducing epithelial-mesenchymal transformation(EMT)process.
