Objective The expression levels of mir-421 in the tissues and plasma of patients with colorectal cancer were measured to compare the differences in the expression of mir-421 in tissues and normal intestinal mucosal tissues.To investigate the correlation between mir-421 and clinicopathology of colorectal cancer and its diagnostic efficacy,and to evaluate its value as a molecular marker for the diagnosis of colorectal cancer.Methods Surgical specimens of 58 colorectal cancer patients and plasma of 30 colorectal cancer patients and healthy subjects were collected.The relative expression of mir-421 in tumor tissues,normal intestinal mucosal tissues and peripheral blood plasma samples was measured by qRT-PCR.The differences in expression levels of mir-421 in tumor tissues and normal intestinal mucosal tissues were compared,and the differences were analyzed in combination with clinicopathological characteristics to explore its correlation with the occurrence and development of colorectal cancer.Meanwhile,the relationship between plasma mir-421 expression levels and clinicopathological characteristics in colorectal cancer patients was compared.ROC was used to evaluate the diagnostic efficacy of mir-421 and to compare it with CEA,a commonly used clinical tumor marker.To evaluate its clinical value as a molecular marker in the diagnosis of colorectal cancer.Results The relative expression level of mir-421 in tumor tissues of colorectal cancer patients was 0.61(0.32,0.86),which was significantly lower than that in normal intestinal mucosal tissues1.31(1.08,1.66)(P<0.05).The difference was statistically significant.In terms of clinical significance,the expression level of mir-421 was not significantly correlated with the age and gender of colorectal cancer patients,but was significantly correlated with tumor size,differentiation,tumor invasion depth,lymph node metastasis and TNM stage(P<0.05),and the difference was statistically significant.The plasma expression level of mir-421 in colorectal cancer patients was 1.70±0.42,significantly lower than that in the healthy group 2.41±0.33,(P<0.05),and the difference was statistically significant.The plasma expression level of mir-421 was not correlated with the patient's age,gender,and TNM stage,but was correlated with tumor size,differentiation,tumor invasion depth,and lymph node metastasis(P<0.05),and the difference was statistically significant..ROC curve results showed that mir-421 was more effective in the diagnosis of colorectal cancer than CEA,a commonly used clinical tumor marker.Conclusions Mir-421 is closely related to the occurrence and development of colorectal cancer.In tumor tissues and plasma of patients with colorectal cancer,the expression level of mir-421 is reduced,and its expression level is related to the clinicopathological characteristics of colorectal cancer.Mir-421 has the potential to become a molecular marker for the diagnosis of colorectal cancer.
