Objective: To detect the miRNAs in cancer tissues,matched adjacent benign colorectal tissues and feces with patients of colorectal cancer(CRC)and to evaluate the diagnostic value of feces miRNA.Methods: Collect clinical specimens who included cancer and matched adjacent benign colorectal tissues and pre-operative feces of 40 cases of CRC who from the first affiliated hospital of Guang Xi Medical University,department of colorectal surgery,and collect feces of 20 cases of healthy volunteers,were enrolled in this study.RNAs were extracted from all this specimens.Test their purity and concentration by electrophoresis and Nanodrop1000.After reverse transcription,Relative expression of a panel of 10 dysregulated miRNAs(miRNA17、miRNA18a、miRNA21、miRNA31、miRNA92a、miRNA96、miRNA143、miRNA181b、miRNA221、miRNA222)were analyzed with QRT-PCR,U6 is the reference genes.Compare the expression of cancer and matched adjacent benign colorectal tissues,feces of patients and volunteers.Elect the obvious miRNAs.Receiver operatingcharacteristic curve(ROC)and area under the curve(AUC)analysis was performed to assess the diagnostic value of fecal miRNAs in CRC patients.Results: The expression of 10 miRNAs in all samples was detected.Compared with matched adjacent benign colorectal tissues,the expression of miRNA-17,miRNA-21,miRNA-31,miRNA-92 a,miRNA-96 and miRNA-221 were significantly higher and the expression of mi R-143 was significantly lower in primary tumor tissues of CRC patients(P<0.01).CRC patients had a significantly higher fecal miRNA-21,miRNA-31,miRNA-92 a and lower miRNA-143 level compared to normal controls(P<0.05).it is positive correlation of miRNA-21,miRNA-31,miRNA-92 a,miRNA-143 between cancer tissues and patient’s feces by correlational analyse.Receiver operating characteristic(ROC)analysis using stool miRNAs expression indicated that Area under the curve(AUC)of combination two of miRNA-21、miRNA-31、miRNA-92 a and miRNA-143 had better than each single.All the combination more than 0.9.While the combination of miRNA-21 and miRNA-143 would be best for detecting and screening CRC with AUC of 0.998,the sensitivity and specificity were 97.5% and 95.3%(P<0.01).Conclusion: Our data indicate that difference of miRNA-21,miRNA-31,miRNA-92 a and miRNA-143 in feces between CRC patient and healthy person were statistical significance.They can become the potential biomarkers of CRC.Combination of miRNA-21 and miRNA-143 may be the no-invasive and,high sensitivity and specificity for diagnosis CRC.
