Periodontal disease is an infectious disease of the oral cavity,which can cause swelling of the gums and destruction of the periodontal tissues,and ultimately the loss of teeth.The ultimate goal of its treatment is to eliminate periodontal inflammation,prevent the occurrence and development of periodontal disease,promote tissue regeneration,and repair the physiological structure and function of periodontal tissue.With the development of oral biomaterials research and the development of various bone tissue regeneration technologies,the effect of bone repair and treatment has been promoted,and the effect of oral implantation technology and clinical implantation repair has been promoted.At present,the research and development of artificially synthesized various implant materials,especially various materials that promote the regeneration of bone tissue,are still the focus of basic research.Objective: The preliminary exploration of the mechanical properties and osteogenesis of calcined fetal calf cancellous bone by hydroxyapatite(HA)and chitosan(CS)surface modification provided new ideas for the study of bone graft materials for periodontal defect repair.Methods: The calcined bone material was calcined by high temperature calcination at two different temperatures and soaked in simulated body fluid(SBF).A large amount of mineralized substances were deposited on the surface of the calcined bone material,and the surface of the calcined bone material was soaked by soaking in the CS solution,which greatly changed the biological properties of the calcined bone surface.Calcined bone material was modified by HA and CS to obtain ?-tricalcium phosphate material(?-TCP),?-TCP/HA material and ?-TCP/HA/CS composite material;then study the characterization of composite materials with EDX spectrum analyzer,universal material tester and Micro-CT;detect the effect of ?-TCP material surface modified by HA and CS on the proliferation and differentiation of rat bone marrow mesenchymal stem cells(BMSCs)with Live-Dead fluorescence kit and MTT kit,Cell adhesion rate experiment was used to study the cell compatibility of ?-TCP surface modified by HA and CS;Enzyme kit and QRT-PCR method were used to detect the expression of alkaline phosphatase(ALP)in early osteoblasts and mRNA of osteogenic marker osteogenic genes in late osteoblasts,and to explore the BMSCs in composite scaffold materials osteogenesis.Results: The main component of calcined bone is ?-TCP,and the composition of mineralized calcined bone surface is HA;the pore size of ?-TCP/HA/CS composite material is 130~446 ?m,and the porosity of ?-TCP,?-TCP/HA and ?-TCP/HA/CS materials are(71.52±0.18)%,(70.86±0.22)% and(70.25±0.22)% respectively,which is no significant change between materials(P>0.05);?-TCP/HA/CS mechanical properties are extremely significantly higher than ?-TCP/HA and ?-TCP material(P<0.01),and ?-TCP/HA is significantly higher than ?-TCP material(P<0.05);all materials are non-toxic and have good compatibility with BMSCs,?-TCP/HA/CS and ?-TCP/HA significantly increased the expression of ALP in BMSCs compared with ?-TCP(P<0.01),and ?-TCP/HA/CS was significantly more significant than ?-TCP/HA(P<0.01);?-TCP/HA/CS is significantly higher than ?-TCP/HA and ?-TCP osteogenes COL?,OPN and OCN(P<0.01);?-TCP/HA and ?-TCP was significantly increased compared to the osteogenic gene COL ?(P<0.01),but OPN and OCN did not change significantly(P<0.05).Conclusion: ?-TCP/HA/CS is a kind of bone grafting material with good mechanical properties,cytocompatibility,and osteogenic induction,which may provide new ideas for the treatment of periodontal tissue defects with dental implant and the development and use of good bone tissue regeneration materials.
