Objective: Compositing Zinc and CBB to get the CBB/Zn2 + composite scaffold materials,in order to improve the mechanical properties and osteogenesis ability of CBB,providing laboratory basis for seeking better bone scaffold material.Materials and Methods: Compositing Zinc and CBB to get the CBB/Zn2 + composite scaffold materials.1)Using XRD,SEM,EDX and INSTRON these universal mechanical instrument testing means,to test the aperture size,porosity,main composition,the content of zinc ions and the mechanical properties of the material.2)Extracting BMSCs from New Zealand in two weeks of immature rabbit,and the extracted cells for osteogenesis and adipogenic differentiation testing,to ensure that the cells of pure stem cells.3)Testing cytotoxicityof materials,observing the adhesion of cells and materials,detecting the expression of ALP and the expression of osteogenesis related gen OCN,OPN and COL?.4)Observing bone defect repair ability of CBB/Zn2+ composite scaffold materials in rabbit mandibular,testing newborn bone mass and degradation rate of the material with Micro-CT,to discusses the osteogenesis ability of the material in the body.Result: 1).The main components of the CBB is ?-TCP,and the zinc content of CBB/Zn2+ is0.32 wt %,the aperture size is 123 ~ 346 ?m,the porosity is about 70% which compared with the CBB has improved but not significant(P > 0.05),while the mechanical properties of CBB/Zn2+compared with the CBB has significantly increased(P < 0.05).2).CBB/Zn2+ belongs to the non-toxic materials and the cells can have a good combine with material.in terms of promoting osteogenesis differentiation,the expression of ALP quantity is improved but not significant(P >0.05),while the expression of osteogenesis related gene improved remarkably(P < 0.05).3).Macroscopic observation,CBB/Zn2+ group repaired area compared with CBB group healed earlier and better,the new bone mass are higher than the CBB group,while the degradation of materials no obvious difference between CBB with CBB/Zn2+.HAP powder group: at 4w,defects of bone healing is the most prominent,and while at 8 w and12 w,because of the large defect in the area,central nutrition in short supply leading to necrosis occurs.Conclusion: CBB/Zn2+ has good biocompatibility,and good mechanical properties of materials,can effectively accelerate the direction of osteoblast differentiation of BMSCs,and promote the secretion of osteogenesis related proteins and related genes,implanted into animals also can promote bone defect repair...
