Study On Cataract Associated With Hypoxic-ischemic Brain Damage In Newborn Mice

Objective:Congenital cataract is a serious blinding lens disease.Studies have found that the risk factors of children born at birth are closely related to the occurrence of congenital cataract.As we all know,perinatal ischemia and hypoxia(Hypoxia-Ischemia,HI)and the resulting neonatal encephalopathy(Neonatal encephalopathy,NE)will affect the vision of full-term neonates.Current research shows that the mechanism of cataracts is related to oxidative stress.According to this,this study constructed a neonatal hypoxic-ischemic brain damage(HIBD)model to simulate neonates caused by perinatal asphyxia Hypoxic-ischemic encephalopathy,after which cataracts,eye development,other eye injuries,and oxidative stress levels in the lens were studied.Methods:C57 mice born 9-11 days were used to ligate the left common carotid artery and placed in an 8%oxygen partial pressure environment to establish the HIBD model.The HIBD model established by this method is the most classic animal model.After successful modeling,observe the eye opening of the mice,and count the eye opening time of the ischemic and contralateral eyes of the model group and the sham operation group,respectively.After opening the eyes of the mice,the table slit lamp was used to observe the lens changes and the pictures were taken.After modeling,the material was taken 3d,and the eyeballs were made into paraffin sections.The HE staining method was used to observe the crystal morphology.Homogenize the lens of the mouse and take the supernatant to detect the activity of superoxide dismutase(SOD),glutathione peroxidase(Glutathione peroxidase,GSH-Px)and Detection of the content of lipid oxide malondialdehyde(Malondialdehyde,MDA).Results:After modeling,the brain damage of HIBD model mice can be divided into:lightly damaged group and severely damaged group.The rate of cataract in the severely injured group is 100%.No cataract was found in the lightly damaged group.Cataracts occurred in the ischemic side of the mice in the severely injured group,but no cataracts were found in the contralateral eyes of the sham-operated group and the lightly damaged group.Over time,the cataracts in the mice persisted and did not progress.In the HIBD severely injured group,the pupils of the eyes with cataracts on the ischemic side of the eyes were dilated and the light reflection was dull.Some cataract mice had increased secretions and corneal opacity in the ischemic eye.Two of the severely injured mice had small eyes,sunken eyes,and limited eyeball development in the ischemic side eyes.The statistical results of the eye-opening time results show that the average eye-opening time of the model group mice is longer than that of the sham-operation group(P<0.05).In the model group,the ischemia-side eye opening time was longer than that of the contralateral eye(P<0.05).Immun The results of HE staining showed that the lens capsule was smooth,the epithelial cells were neatly and densely arranged,and the collagen fibers were neatly arranged in the sham operation group.Both the severely and lightly damaged groups had rough lens capsules,the epithelial cells were disorderly arranged,and the collagen fibers were disorderly arranged,but light The damaged group had less damage than the severely damaged group.Detection results of oxidative stress kinase:The MDA content in the lens of the ischemic side of the mice in the three groups was significantly higher in the severely damaged group than in the mildly damaged and sham-operated groups.There was a statistically significant difference between the two groups(P<0.001).Compared with the two groups,the content of MDA in rats with ischemic lateral eyes and contralateral eyes was significantly increased(P<0.001);the SOD and GSH enzyme severely damaged groups were significantly lower than those in the two groups,and the comparison between the two groups was statistically significant(P<0.005).In the model group,the enzymatic activity of the ischemic eye was significantly lower than that of the contralateral eye(P<0.005).Conclusions:1.Congenital cataract occurred in the ischemic lateral eye of the HIBD model severely injured group.2.The average eye opening time of mice in the HIBD model group was significantly prolonged.The eye opening time of the ischemic side was longer than that of the contralateral eye.The eyeball development was restricted,the pupils were dilated,and the effect of ischemia and hypoxia on eye development was more.Aspects.3.Oxidative stress occurred in the lens of ischemic eye of HIBD model mice,antiperoxidase activity decreased,and the level of oxidative stress was related to the occurrence of congenital cataract.
