Boschniakia rossica(Cham.Et Schlecht.)Fedtsch is a plant belonging to the genus Ranunculaceae.Caorongrong is a folk medicine,mainly used to treat kidney deficiency,impotence,cold pain in lumbar joints,constipation,etc.Boschniakia rossica contains a variety of biological activities such as anti-aging,anti-tumor,anti-inflammatory,immune system regulation,anti-fibrosis,hepatoprotective effect and memory improvement.Therefore,in this thesis,the main constituents of Boschniakia rossica are analyzed and identified by HPLC-MS.The HPLC method is used to quantitatively analyze the two major components.Based on this,a combination of liquid chromatography.The main components were separated and extracted;the effects of Boschniakia rossica extract on hydrocortisone-induced kidney yang deficiency in a mouse model were explored.Research indicates:1.Preparative liquid Chromatography phase separation technology was used to prepare the main active ingredients in Boschniakia rossica.The isolated compounds were determined by 13C spectrum and spectrum to determine the isolated compounds were identified as Boschnaloside and Rossicaside B.2.HPLC was developed for the simultaneous detection and analysis of rushin nalaraside and rushin Method of asterinoside B:Column Klimail 100-5 C18(250 mm × 4.6 mm,5 ?m);mobile phase:acetonitrile(A)-0.05%formic acid water(B);flow rate:1 mL/min;Temperature:30?;detection wavelength:260 nm;injection volume:20?L;gradient elution conditions:0-12 minutes,15%(A);12-30 minutes,15-20%(A);30-40 Minutes,20-25%(A);40-45 minutes,25-30%(A);45-60 minutes,30-100%(A).The results showed that the method was sensitive,accurate and stable,and provided a method for the quantification of Boschnaloside and Rossicaside B in cistanche.3.In this study,a large-dose intraperitoneal injection of hydrocortisone induced an animal model of Kidney-Yang deficiency in mice.Compare the effects of Boschniakia rossica extract with high,medium and low doses and oral administration of Jinkui Shenqi Pills on kidney yang deficiency:The serum SOD and MDA contents were used as indicators to investigate the dynamic balance of oxygen free radical reaction and lipid peroxidation reaction;the contents of T,FSH,cAMP and cGMP in serum were measured,and the changes of sex hormones in animals were measured;ActR2A and BMP-5 content to investigate changes in animal bones.The results showed that the Jinkui Shenqi Pill and Boschniakia rossica high-dose group can significantly increase the SOD activity,decrease the MDA content,significantly increase the serum T and cAMP levels,decrease the cGMP content,and decrease ActR2A and BMP-5 in kidney-yang deficiency mice content increased,kidney yang deficiency proved to be significantly improved.In summary,we established HPLC for simultaneous detection and analysis of two main compounds in the plant(Boschnaloside and Rossicaside B);Isolation and extraction of the main components of Boschniakia rossica;analysis of therapeutic effects of Boschniakia rossica extract on hydrocortisone-induced kidney-yang deficiency mice.This study provided theoretical basis and scientific support for the development and utilization of Boschniakia rossica.
