Inflammation is a kind of defence reaction,which can react when the tissues with blood-vascular system in living organisms are damaged by various kinds of causative agents. It is characterized by the activation of leucocytes and endotheliocytes. Inflammation is very common and important .it participate in many basic pathologic processes, such as bacterial infection, wounds,tumer ,artherosclerosis and some other immunolgically mediated diseases. So it is thought the common ground of many diseases. At present, the known anti-inflammatory drugs can be divided into three types: non-steroid anti-inflammatory drugs (NSAIDs),steroid anti-inflammatory drugs,(SAIDs), disease modifying drugs (DMDs).Though they all have anti-inflammatory effect,some of them have side effect.Therefore it is increasingly important to develop a kind of effective and harmless anti-inflammatory durg.Polysaccharide is natural high polymer which comes from cellular membrane of advanced plants and animals. It also exists in the cell wall of microbes. It takes part in the vital movement togethering with protein,fat and nucleinic acid.Along with chemistry and biology improving rapidly and isolation technics developing , people have different cognition about Polysaccharide gradually. We don't think it is the energy source and tissue components only any more.People in growing numbers are beginning to pay more attention to the Polysaccharide due to its pharmacological action. At present, Polysaccharide as a pharmacological research direction, its anti-inflammatory effects is becoming the hot spot in the research field. Boschniakia rossica is a kind of rare medicinal plant which mainly distribute in the Changbai Mountain area in China. PBR is the polysaccharide extracted from Boschniakia rossica, it has higher medicinal properties and broad exploitation prospect . In recent years,some literatures report that it enhances the proliferative response of B lymphocyte to lipo-polysacharides, have mitogensis to splenocytes and improve immunity in mice and so on. This article studies the PBR anti-inflammatory reaction systematically and its mechanism initially in order to undergo this research indepth.In this experiment,we establish three experimental models. One is making the ear of mice edema by dimethylbenzene , another is causing the hindpaw of the rat edema by dextran,the last one is acute peritonitis of the mice. Later we use biochemistry analysator to detect the variances of leukocytes and neutrophils. This can help us research the PBR anti-inflammatory reaction. At the same time it reveals the mechanism of anti-inflammation inatially. We also evaluate the safety of PBR by acute toxicity test , maximum tolerated dose test , micronucleus test of bone marrow PCE cells ,sperm abnormality test. The result shows that PBR markedly inhibited ear edema induced by dimethylbenzene,0.4 g/kg and 1.6 g/kg dose PBR has strong inhibition (P<0.01).PBR inhibit leukocytes and neutrophils of blood of mice ear edema induced by dimethylbenzene significantly , the effect between 0.4 g/kg and 1.6 g/kg appears obviously , compared with the PBS group (P<0.01). PBR markedly inhibits hindPaw edema induced by dextran in rats(P<0.05),the effect of 140 mg/kg PBR is most effective(P<0.01).Its effect surpass 280mg/kg does(P<0.05). 0.4 g/kg and 1.6 g/kg dose PBR has strong inhibition to mice acute peritonitis (P<0.01) , it demonstrates dose-dependent . Experimental results show that PBR has significant anti-inflammatory effect. The toxicity test of PBR shows LD 50 > 24 g/kg , the maximum tolerated dose test shows MTD > 30 g/ kg, the micronuclear rates of low, middle and three high-dose group are 2.2‰,2.9‰and 2.3‰, the distinction is outstanding compared with the positive group(P<0.01). the sperm abnormality rates of low, middle and three high-dose group are 2.3 %,2.0 % and 4.0 % respectively, the discrimination is prominent, compared with the PBS group(P<0.01). Experimental results show that PBR is non-toxic and can not induce the micronuclear rates and sperm abnormality rates of mice increasing.It indicates that PBR is asepsis and non-mutagenic effects . PBR can be use it safely ..
