Objective:To explore the mRNA and protein expression levels of sperm associated antigen 5(SPAG5)in gastric cancer(GC)tissues,and evaluate its correlation with the clinical-pathological characteristics of patients and the prognostic value.Methods:The clinical data of 41 patients who underwent radical gastrectomy in First People's Hospital of Changzhou from September 2015 to March 2017 were collected and gastric cancer tissue microarray consisting of 90 cases was applied.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expression of SPAG5 mRNA in 41 cases of gastric cancer tissues.Immunohistochemical technique was used to detect the expression of SPAG5 protein in gastric cancer tissues and commercial gastric cancer tissue microarray.The rank sum test was used to compare the difference of SPAG5 expression in gastric cancer tissues and adjacent tumor tissues.The correlation between the expression of SPAG5 and clinicopathological characteristics of gastric cancer was analyzed by the Mann-Whitney U test or ?2 test and Fisher exact test.Kaplan-Meier method was adopted to survival analysis and plotted the survival curve.The survival differences between groups were analyzed by the Log-rank test.Results:SPAG5 mRNA was detected in all 41 cases of gastric cancer tissues,the median relative transcript level was 0.300(0.145,0.683).There was no correction between the expression of SPAG5 mRNA with age,sex,tumor stage,lymph node status,degree of differentiation,depth of tumor invasion and tumor size(all.P>0.05);75.6%(31/41)had a high level of SPAG5 protein and 24.4%(10/41)had a low level.There was also no correction between the level of SPAG5 protein with age,tumor stage,lymph node status,degree of differentiation,depth of tumor invasion and tumor size(all P>0.05),but there was significantly correlated with sex(P=0.039),and the level of SPAG5 in female patients was higher than that in male patients.Survival analysis showed that the level of SPAG5 mRNA and protein expression was not significantly correlated with disease-free survival(DFS)and overall survival(OS)of patients(all P>0.05).The expression level of SPAG5 was significantly higher than that of adjacent tissues in gastric cancer tissue microarray(P<0.001),but there was also no significant correlation with patient age,sex,degree of differentiation,tumor stage,depth of tumor invasion,lymph node status,tumor size and overall survival(all P>0.05).Conclusion:SPAG5 expression was detected in all gastric cancer tissues and its expression level was significantly higher than the adjacent tissues.The expression of SPAG5 protein may be related to the gender of patients.SPAG5 has no prognostic value on gastric cancer patients.
