Objective To investigation the expression of sperm associated antigen5(SPAG5)and microtubule associated protein l light chain 3(LC3)in primary breast invasive ductal carcinoma,and discusse the relationship between expression,clinicopathologic characteristics and sensitivity to neoadjuvant chemotherapy in breast tumor.Methods The paraffin specimens of tissue from 107 patients with primary breast invasive ductal carcinoma were collected before and after TEC regimens(T :Docetaxel E: Epirubicin C: Cytoxan)neoadjuvant chemotherapy,SPAG5 and LC3 expression were evaluated by immunohistochemical staining.Data were analysed to investigate the correlation to clinicpathologic factors and chemotherapy response.Results The high-expressed rate of SPAG5 from tissues of breast tumor was 48.6% and proved to be positively correlated to the over-expression of proliferating cell nuclear antigen(P = 0.019),LC3(P=0.013)and human epidermal growth factor receptor-2(P=0.000).The over-expressed rate of LC3 was 67.2%,and proved to be positively correlated to the over-expression of human epidermal growth factor receptor-2(P=0.002)。There were no significant correlations between age,tumor size,TNM stage,histological grading,lymphnode status and the expression level of estrogen receptor and progesterone receptor.The overall objective response(OR)rate was 70.1% and the pathologic complete response(p CR)rate was 9.3% after neoadjuvant chemotherapy.The objective response rates were 82.7% in SPAG5over-expression group significantly higher than 56.1% in low-expression group(P=0.006).The rates of p CR were 15.4% group of SPAG5 over-expression significantly higher than 3.6% in low-expression group(P=0.048).The OR rate and p CR rate of patients with over-expression of SPAG5 were higher than low-expression cases.The OR rates were 76.4% in LC3 strongly positive expression group higher than54.3% in low-expression group(P<0.05).That is no statistically significant between LC3 expression and p CR rate.Conclusion SPAG5 as a tumor proliferation index,its expression status for breast cancer patients with TEC regimen neoadjuvant chemotherapy has a predictive effect.
