Objective:In this study,the SD rat model of Parkinson's disease and Control group were constructed by using different drugs.The aim is to research the expressions of p75 and siAH in these two different groups.From our study,we can go further to explore pathogenesis of Parkinson's disease,providing guidance for future clinical treatment and improve the long-term prognosis of patients.Methods:A total of 50 Sprague Dawley rats with 230-280g were divided into two groups called Control and Rotenone were used in this study.We ensure the consistent conditions in the feeding environment.The model was constructed using the brain stereotactic method.After the model matured,the apomorphine induction test was used to verify at the 4th week.The rats with successful modeling were sacrificed to get the related brain tissue.But there have different methods to do.For rats which need to be studied through the immune fluorescence,brain tissue perfusion was used to fix and slice.By adding a blocking solution and a primary antibody to the brain slice,the green and red light double labeling and nucleation were performed with a fluorescent secondary antibody and a nuclear nucleus reagent,and the expression of the target protein was observed under an immune-focus microscope.For Real-time PCR and Western-blot group,the rats were taken out of the fresh tissue on ice and immediately placed in liquid nitrogen for storage.For Real-time PCR,the specific operation method is carried out according to the instructions.And when performing the Western-blot experiment,the protein is extracted firstly,then the concentration is measured,and then the steps are followed by mixing,electrophoresis,membrane transfer,antibody addition,and finally exposured.Finally,the relevant pictures and data are obtained,the statistical analysis of the pictures and data is carried out,and the experimental conclusions are summarized.Results:(1)After the model was established in these two groups,there was no statistically significant differences in weights,which indicates that the weights changes of rats in Control and Rotenone group were basically the same;(2)In the fourth week with the induction of morphine(APO),the number of rotations in the Rotenone group was significantly higher than the other group,which indirectly proved that the Parkinson's model induced by Rotenone was meaningful;(3)Compared with the Control group,the expression level of TH assay decreased significantly in the Rotenone group.Besides,the expression levels of p75,Ub and siAH in Rotenone group increased with different degrees,indicating that p75 and siAH both played an important role in the publication of Parkinson's disease.At the same time,these results provide deeper clinical thoughts to clinical treatments.
