Objective: Intraperitoneal injection of LPS-induced Parkinson’s disease model of C57 BL / 6 mice to investigate the behavioral changes of PD model mice and the presence and metastasis of a-synuclein aggregates in the intestinal wall and the dopaminergic nerve Damage to the damage situationMethods: 1.C57 BL / 6 mice were randomly divided into blank control group and PD model group.PD model group: injection of lipopolysaccharide(LPS)(5mg per kilogram of body weight,dissolved in saline),only the first day of the experiment by intraperitoneal injection;blank control group: normal saline intraperitoneal injection,the same time and dosage Two groups.2.The exercise coordination ability of the mice was observed by fatigue experiment with 1 month,3 months,5 months and 7 months respectively.The exercise ability and spatial exploration behavior of the mice were evaluated by open field experiment.3.The expression of TH,a-synuclein in the intestine of the intestine and midbrain substantia nigra was detected by immunohistochemistry.The relative contents of TH-a synuclein were detected by Western blotting Western blot was used to determine the expression levels of P-JNK,P-P38 and P-ERKResults: 1.Intraperitoneal injection of LPS can significantly reduce the number of dopaminergic neurons in the substantia nigra of mice 2.A-synuclein was first expressed in the intestinal nerves of Parkinson’s disease mice and gradually appeared in the midbrain substantia nigra shortly as time prolonged..3.LPS can turn bar Parkinson’s disease on fatigue in mice experiment tolerance time shorten and reduce the number of standing in kuang field,the number of times and through the grid.4.LPS could affect the activation of MAPK pathway in Parkinson’s disease model mice: Compared with the control group,the expression of P-JNK and P-P38 in the model group was significantly higher than that in the control group(P <0.01)There was no significant difference in the level of P-ERK(P> 0.01)Conclusion: 1.Intraperitoneal injection of LPS can successfully produce Parkinson’s disease mouse model,so that Parkinson’s model mice coordination ability,athletic ability and spatial exploration behavior decline,the mechanism may be associated with chronic inflammation of the brain,MAPK pathway activation.2,LPS,the coordination of the Parkinson’s disease model mice can be reduced,athletic ability and space exploration.2.In the LPS-induced PD model,the typical pathological changes(α-Syn agglutination)may initially be expressed in the colon nerves and progress gradually to the midbrain substantia nigra 4,intraperitoneal injection of LPS can lead to PD model mice midbrain dopaminergic neuron injuries of the substantia nigra.3.Intraperitoneal injection of LPS could lead to dopaminergic neuronal damage in substantia nigra of PD model mice...
