Objective: The research of the quality control on eucommia ulmoides Oliv of Guizhou aims at examining its quality stability by the method of the HPLC and HPCE fingerprints of it, exploring the new method of controlling the quality of it, and providing some experimental evidences for the foundation of the quality standard of it.Methods: The research chooses 12 batches of eucommia ulmoides Oliv from different parts of Guizhou to examine the influence of different extractions and rates of solvent and medicine material on distilling the effective ingredient from eucommia ulmoides Oliv by optimizing the beforehand handling procedure of quality control through experiment which focuses on three major factors affecting the efficiency of extraction, including the extraction solvent, extraction time and extraction times.The experiment adopts HPLC and HPCE to analyze the fingerprints of eucommia ulmoides Oliv, introduces 2004A edition of the evaluation system on similitude degree of chromatogram fingerprint of chinese traditional medicine compiled by the State Pharmacopeia Committee of China to evaluate the HPLC and HPCE fingerprints of eucommia ulmoides Oliv, chooses no less than 10 batches of representative eucommia ulmoides Oliv to establish the common mode of the HPLC and HPCE fingerprints of eucommia ulmoides Oliv, and analyzes the similitude degree of eucommia ulmoides Oliv coming from different parts of Guizhou to examine the stability of Guizhou's eucommia ulmoides Oliv. It analyzes the HPLC fingerprint, meanwhile, establishes the method of quantity testing of the effective components in eucommia ulmoides Oliv. It also use the new method to test the quantity of Chlorogenic acid in eucommia ulmoides Oliv by adopting the internal standard method to make capillary zone electrophoresis.The experiment also makes the representative eucommia ulmoides Oliv salted according to the salt making method in the 2005 edition of Chinese Pharmacopoeia, analyzes the salted medicine by HPLC fingerprints to establish the common mode of HPLC fingerprints, and reflects the changes of chemical components before and after the salt making by comparing their respective common mode of HPLC fingerprints.Result: The experiment ascertains that the beforehand handling procedure of quality analysis of eucommia ulmoides Oliv is the ultrasonic extraction of the mixture of five portion of 60% ethanol and one portion of wadding powder of eucommia ulmoides Oliv, which are performed once and lasts for 30 minutes.It establishes the investigating method of analyzing the HPLC fingerprints and testing the components quantity simultaneously. It find out that the similitude degree of two out of the 12 batches of Guizhou's eucommia ulmoides Oliv is under 0.9. Then it evaluates the similitude degree of the other 10 batches of eucommia ulmoides Oliv and its salted ones, and finds out that the similitude degree of eucommia ulmoides Oliv is between 0.904 and 0.989, and the similitude degree of its salted ones is between 0.904 and 0.999. It also establishes the common mode of the HPLC fingerprint of the 10 batches of eucommia ulmoides Oliv and their salted ones and confirms that they have 19 common apexes. The quantity of Pinoresinol diglucoside and Chlorogenic acid in eucommia ulmoides Oliv are 1.042~4.47mg·g-1 and 0.053~2.435mg·g-1It also upbuilds the research method of HPCE fingerprints of eucommia ulmoides Oliv and the quantity testing method of Chlorogenic acid in eucommia ulmoides Oliv, and concludes that the similitude degree of the HPCE fingerprint of the 10 batches of Guizhou's eucommia ulmoides Oliv is between 0.903 and 0.991.Conclusion: The analyzing method of HPLC and HPCE fingerprint of eucommia ulmoides Oliv have the features of easy operation, high exactitude and excellent repetition, both of which can be used to control the quality of eucommia ulmoides Oliv. The majority of Guizhou's eucommia ulmoides Oliv have stable quality which can be controlled, while some of them have diverse quality which need to be further investigated. The analyzing method of HPCE is simple, convenient, speedy and economical, which makes up for the disadvantage of HPLC in some degree and can be regarded as one new method to control the quality of eucommia ulmoides Oliv.
