Aging is an inevitable tendency in life, and the aging in human is usually accompanied by the occurrence of senile diseases such as arthritis, diabetes, high blood pressure, Alzheimer's Disease,cataracts, heart disease etc. The process of aging is sophisticated, which are affected by many factors. Longevity is the extreme purpose of anti-aging approach. Although many genetic modifications indicated longevity, calorie restriction still remains to be the only robust external intervention across variety of species. Therefore, scientists keep the motivation to approach the medicine related longevity. With the development of science and technology in chemical monomer, the molecular mechanism of the aging process will be eventually clarified.. Therefore, more and more attention has been focused on the research of anti-aging medicines.With the continuous development of Chinese medicine, The treatment of traditional Chinese medicine is reported as unique curative effect and less side effect, which is widely used in the treatment of aging related miscellaneous diseases. One of the anti-aging herbs is the tonic herb, Eucommia ulmoides, which is one of clinical common toning herbs has function of tonifying liver and kidney, strength the bones and muscles, lowing the body-weight and be resistance to aging. According to the modern pharmacology, the effects of Eucommia ulmoides included the liver protection and cholagogue, antioxidant and anti-aging effect.This article applied C. elegans(Caenorhabditis elegans) as a model organism to study an active component in Eucommia ulmoides as the primary study on the mechanism of delay aging. The research content and result is as follows:Determination of the water extract of Eucommia ulmoides, and the monomers such as aucubin, genipin geniposide acid effect on the life cycle of C. elegans. Base on the significant lifespan extension effect, the study in heat shock resistance, antioxidant ability damage, spontaneous mobile ability, dynamic swing frequency, swallowing ability, lipofuscin deposition of geniposidic acid is also performed.1 The lifespan(survival rate) of C. elegans in Eucommia ulmoides water extract, aucubin, geniposide acid were analyzed with the congeneric strain of wild type fem-1(hc17) Geniposidic acid treatment was demonstrated to improve the average life expectancy to 15%.2 Investigate the effects of geniposidic acid on the related physiological indexes of anti-aging effect of C. Elegans fem-1(hc17) such as heat shock resistance, antioxidant ability damage, locomotor activities and mobility etc.3 Geniposidic acid has been demonstrated to improve the heat shock resistanceability of fem-1(hc17). Under 35 oC, 1×10-3mmol/L Geniposidic acid treatment for 8 hours, the survival rate has increased the 42% to 58%(blank vs treatment), which improved the survival rate of 38%. At 10 hours, the survival rate has increased from 23% to 43%(blank vs treatment), which improved the survival rate of 87%.4 The resistance to oxidative damage of the C. elegans by hydrogen peroxide has also increased significantly, and the survival rate was increased.When the concentration of H2O2 was 0.002%, the survival rate of the C. elegans 12 h in the treatment group was increased to more than 75%. Each concentration had better antioxidation effect.5 The spontaneous movement ability of the nematode was promoted by the action of the geniposidic acid, and it could significantly increase the C. elegans swing frequency, increase the activity ability of the nematode, and had a positive effect on the muscle function.6 After the treatment with geniposidic acid for 5 days and 10 days, the Pharynx-pumping rates of the treatment group was significantly increased. This result indicated a significant improvement of the swallowing muscle of the C. elegans, which indicated the requirement of muscle function of the anti-aging effect of geniposidic acid.7 After 15 days treatment with geniposidic acid, the accumulation of oxidative cross-linking protein lipofuscin was lower than the blank group 20%, which related with a lower fluorescence density. This result may indicate the reducing of lipofuscin deposition caused by anti-aging of geniposidic acid.In summary, The geniposidic acid can delay the aging in C. elegans and the anti-aging mechanism has a close relationship with the increase in muscle tone.
