Medullary thyroid carcinoma is a relatively rare type of thyroid tumor.It has the characteristics of high malignancy,rapid development,and poor prognosis.Currently,surgery,radiotherapy,chemotherapy are mostly used for treatment,but it seems that all of them are not so effective.The treatment and prognosis of patients with medullary thyroid cancer are related to the time of whom were diagnosed.The sooner whom were diagnosed,the cleaner the tumor is excised by surgery,the better the prognosis of the patient.Therefore,how to detect and diagnose medullary thyroid cancer early is of great significance for patients.As one of the tumor markers of medullary thyroid carcinoma,calcitonin,which can play a very important role in assisting diagnosis for MTC.Although calcitonin can assist the diagnosis of medullary thyroid cancer,but many diseases,such as small-cell lung carcinoma,acute or chronic renal insufficiency,renal failure,gastrointestinal cancer,breast cancer and carcinoid syndrome can elevate the level of serum calcitonin,and it still has shortcomings such as short half-life and so on.Procalcitonin as a pre-peptide substance of calcitonin,there has some cases of abnormally elevated Procalcitonin of medullary thyroid cancer were reported in some literatures.However,there is no relevant basic research on whether procalcitonin can be used as an auxiliary diagnostic marker for medullary thyroid cancer and the relationship between procalcitonin and calcitonin.Objective:By establishing a series of human medullary thyroid carcinoma model which were based on mice,the relationship between the level of serum calcitonin and procalcitonin and the tumor size were studied.And we also analysis the correlation between the level of serum calcitonin and procalcitonin,just because of the serum calcitonin was used as the tumor marker of the human medullary thyroid carcinoma.The study of human thyroid carcinoma based on mouse model provides some experimental evidence for procalcitonin as a diagnostic marker of human medullary thyroid carcinoma.Methods:30 mice were divided into 3 groups,two experimental groups which were marked group A and group B,and a control group marked as group C.10 mice for each group,and each group was equally divided into male and female.The mice of group A and group B were intraperitoneal injected by 80mg/kg BW/d of cyclophosphamide to achieved immunosuppression.Then 2×107 per milliliter of medullary thyroid carcinoma TT cells per mouse to inoculate subcutaneously with the mice of group A which were achieved immunosuppression after intraperitoneal injection of cyclophosphamide.The mice of group B were injected with an equal volume of normal saline.The control group C did not intervene.When the mice of group A were successfully tumor-bearing,measured the length and width of the tumors daily.After the minimum width of the tumor is wider than 0.5cm,the mice in the experimental group A and B,the control group C could be executed and the serum was collected for detecting the serum calcitonin and procalcitonin.The tumors of the mice in the experimental group A were stripped and used for pathological diagnosis.The Measurement data of mouse body weight,mouse tumor weight,tumor volume,mouse serum calcitonin level and procalcitonin level were carefully recorded and expressed as(?)± s.The statistical analysiss was performed by SPSS 22.0 software for One-Way Analysis of Variance which was used to compare the data of mice between group A,group B and group C.The paired-samples t-test which was used for the comparison between data of the same groups before or after some intervention factors.And the Pearson test was used for the correlation of variable data.The test level was a=0.05,and P<0.05 was statistically significant.Results:1.The tumor formation rate of mice in the experimental group was 100%and all of these mice were alive during the experiment.2.At the beginning of the experiment(in the first week),the body weight of the experimental group A and B and the control group C had no significant statistical difference in the body weight of the mice(p>0.05).And when in the last week of the experiment(in the fourth week),the body weight of the experimental group A and group B had a significant difference(p<0.05),the experimental group A and the control group C also had a significant difference(p<0.05),but when group B and the control group C were compared,there had no significant difference(p>0.05).3.The serum calcitonin level of experimental group A and group B and control group C was statistically significant(P<0.05);There had a statistically significant difference in serum procalcitonin level between experimental group A and group B.And experimental group A and control group C also had a statistically significant difference(p<0.05).But when group B and the control group C were compared,there had no significant difference(p>0.05).4.The body-weight,tumor volume,tumor weight,serum calcitonin level,and procalcitonin level in the experimental group A were positively correlation,and the correlation R values are(R1=0.995,R2=0.930,R3=0.937,R4=0.708;P<0.05);the serum calcitonin level and procalcitonin level were also positively correlated and the correlation R value is(R5=0.911,P<0.05).After controlling the variable of body-weight of mice in the experimental group A,the tumor volume,serum calcitonin level,and serum procalcitonin level were still significantly positively correlated with tumor weight in the experimental group A and the correlation r values are(r1=0.990,r2=0.889,r3=0.892;P<0.05).And the serum calcitonin level and procalcitonin level were also positively correlated and the correlation r value is(r4=0.855,P<0.05).5.Pathological experiment also confirmed that the tumor cells which were bearing on the body of mice in the experimental group A was medullary thyroid carcinoma TT cells.Conclusions:1.Successfully established a tumor model of human thyroid medullary carcinoma cells implanted subcutaneously in immunosuppressed mice.Because this model not only has an in vivo immune environment similar to that of tumor patients,it also has a similarity on the pathological characteristics to the human medullary thyroid carcinoma.The experiment also provide a new animal model for future research on human medullary thyroid cancer.2.There has a significant positive correlation between the level of the serum procalcitonin and the tumor weight.And there also has a significant correlation of serum calcitonin and procalcitonin.That means the serum procalcitonin can also be used as a tumor marker for medullary thyroid cancer,just like the same meaning of serum calcitonin for the diagnosis of medullary thyroid cancer.The procalcitonin may also have application value for the diagnosis of medullary thyroid cancer.
