Expression Of CST2 And CENPI In Gastric Cancer Organization And Their Correlation With Clinicopathology

Objective: The purpose of this study is to explore the expression of CST2 and CENPI in gastric cancer tissues and adjacent tissues and their correlation with clinicopathology in patients with gastric cancer,in order to screen new tumor markers of gastric cancer and provide a new direction for the diagnosis and treatment of gastric cancer.Methods: In this study,using UALCAN analysis of the portal of TCGA database,we found that cystatin-SA protein gene(CST2)and centromere protein I gene(CENPI)may be related to the occurrence and development of gastric cancer.The specimens of gastric cancer and paracancerous tissues of 30 patients with gastric cancer admitted to our hospital from January 2017 to March 2019 were collected.Real-Time PCR was used to detect the expression of CST2 and CENPI in gastric cancer tissues and paracancerous tissues,and the experimental data were statistically analyzed.Using the clinicopathological data of patients with gastric cancer in TCGA database,the relationship between the expression of CST2 and CENPI in gastric cancer and the clinicopathology of patients was analyzed.The clinicopathological data of 30 patients with gastric cancer were collected and statistically analyzed from six aspects: age,sex,degree of differentiation,lymph node metastasis,tumor T stage and TNM stage,to further verify the relationship between the expression of CST2 and CENPI in gastric cancer and the clinicopathology of the patients.Results: The analysis of portal UALCAN using TCGA database showed that the expression of CST2 in gastric cancer tissues was significantly higher than that in adjacent tissues,which may be the prognostic risk gene of gastric cancer.CENPI expression in gastric cancer tissues is significantly higher than that in adjacent tissues,which may be the oncogene of gastric cancer.The gene expression was detected by Real-Time PCR and the difference of expression was analyzed.Compared with the matched paracancerous tissues,the expression of CST2 was high in 26 cases and low in 4 cases.The difference was statistically significant(P < 0.01),indicating that the expression level of CST2 in gastric cancer tissues was significantly higher than that in paracancerous tissues.Compared with matched paracancerous tissues,the expression of CENPI in gastric cancer was high in 21 cases and low in 9 cases.The difference was statistically significant(P<0.05).The results showed that the expression level of CENPI in gastric cancer was significantly higher than that in paracancerous tissues.Furthermore,using the clinical data of 327 patients with gastric cancer with complete clinicopathological data in TCGA database,the results showed that the expression level of CST2 was correlated with TNM stage(P<0.05),but not with age,sex,T stage,distant metastasis and lymph node metastasis(P >0.05).The expression of CENPI was correlated with gender(P<0.05),but not with age,T stage,distant metastasis,lymph node metastasis and TNM stage(P > 0.05).According to the results of Real-Time PCR test and the clinicopathological data of 30 patients,it was concluded that there was no correlation between the expression of CST2 and age,sex,T stage,differentiation degree,lymph node metastasis and TNM stage of gastric cancer patients(P > 0.05).There was no correlation between CENPI expression and age,sex,T stage,differentiation,lymph node metastasis and TNM stage in patients with gastric cancer(P > 0.05).Conclusion: CST2 and CENPI are significantly overexpressed in gastric cancer tissues and are involved in the development of gastric cancer.Among them,CST2 may be related to the stage TNM gastric cancer patients,and CENPI may be related to the sex of gastric cancer patients.
