| Objective: To observe the clinical efficacy of Huayu Qushi Decoction in treating patients with blood stasis and damp stagnation type knee osteoarthritis,as well as the changes of serum IL-1,MMP-1,MMP-3 and TIMP-1 levels in patients before and after treatment.To explore its therapeutic mechanism in terms of clinical efficacy and micro-cell level.Methods: Forty patients who had been diagnosed with knee osteoarthritis in the First Affiliated Hospital of Guangxi University of Traditional Chinese Medicine from May 2019 to November 2019 were eligible for inclusion.Randomized controlled clinical trial research methods were used to randomly divide the cases into 2 groups(random number table method),namely,the Huayu Qushi Fang test group and the Irexibu tablet control group,with 20 cases in each group.There was no statistical difference between the two groups in gender,age,duration of disease,and number of single and double knees.The course of treatment is 2 weeks.Before treatment and 2 weeks after treatment,TCM syndrome scores and pain visual simulation scores were used to comprehensively evaluate the two groups of patients to determine clinical efficacy,and 2 weeks after the treatment was completed,the TCM syndrome score was used to score again Efficacy;Patients' serums were collected before and 2 weeks after treatment to test the changes of IL-1,MMP-1,MMP-3 and TIMP-1 levels to observe the effect of drugs on serum cytokines.The research data was analyzed by SPSS22.0 software.Results: 1.Clinical research: The Huayu Qushi Decoction test group and the Airexibu tablet control group achieved clear results in the treatment of knee osteoarthritis.(1)After treatment,the differences in TCM syndrome scores and pain visual analogue scores between the two groups were statistically significant(P <0.05),indicating that Huayu Qushi Fang has a significant effect in treating KOA and is superior to the control group;(2)trial The incidence of adverse reactions in the group(10%)was lower than that in the control group(20%),but the difference between the groups was not statistically significant(P> 0.05);(3)The effective rate of the test group was 95%,and the effective rate of the control group was 65%.The experimental group was better than the control group,and the difference between the groups was statistically significant(P <0.05);The effective rate was 80% and the control group was 55%.The experimental group was better than the control group,but the difference between the groups was not statistically significant(P> 0.05).2.Cytokine research: Serum IL-1,MMP-1,and MMP-3 were significantly lower in the two groups after treatment than before treatment(P <0.05),and TIMP-1 levels in the two groups were significantly higher than those after treatment.Previously,the hint may be related to the two groups being able to inhibit the production of IL-1,MMP-1,MMP-3 and increase the production of TIMP-1.(1)After treatment,the levels of IL-1 and MMP-1 in the test group were significantly lower than those in the control group,and there was a significant difference between the groups(P <0.01),indicating that the test group was superior in inhibiting the production of IL-1 and MMP-1.Control group;(2)MMP-3 levels in the test group after treatment were lower than those in the control group,but there was no significant difference between the groups(0.01 <P <0.05);(3)TIMP-1 levels in the test group were significantly higher than those in the control group after treatment.There was a significant difference between the two groups(P <0.01),indicating that the test group was better than the control group in promoting TIMP-1 production.Conclusions: Huayu Qushi Decoction is effective in treating blood stasis and dampness type Knee osteoarthritis and has fewer side effects.It can reduce the abnormally elevated levels of IL-1,MMP-1,and MMP-3 in patients and increase the level of TIMP-1,improving patients Quality of life.Huayu Qushi Decoction can achieve certain therapeutic goals by regulating the above indicators. |
