Experimental Study Of Zhongtong Fumigant Lotion On Nf-?B P65 And Related Inflammatory Factors Of Pain Model Rats After Anorectal Surgery

Objective: To investigate the effect of Zhongtong fumigant lotion on the expression of NF-?B p65 protein in anorectal tissues and the levels of serum IL-6,COX-2 and TNF-? of anorectal pain model rats after anorectal operation,and to study the mechanism of Zhongtong fumigant lotion in anti inflammation and analgesia after anorectal operation.Methods: 42 healthy and clean SD male rats were randomly divided into 6 groups,7 rats in each group,which were blank control group(normal group),model group,normal saline group,Musk Hemorrhoids Ointment group(hemorrhoids ointment group),Zhongtong fumigant lotion group(fumigant lotion group),Zhongtong fumigant lotion combined with Musk Hemorrhoids Ointment group(combined medication group),all of which established anorectal diseases after operation except the normal group Pain model,the combination group and the fumigant group were used to fumigate and wash the anorectal wounds,the normal saline group was used to fumigate and wash the wounds,the combination group and the Hemorrhoid Cream group were used to local smear the wounds(the above groups were used twice a day for three days),the model group and the normal group were not treated.Three days later,the heart blood was collected and preserved.The contents of IL-6,COX-2 and TNF-? in serum of rats in each group were measured by enzyme-linked immunosorbent assay(ELISA).The anus rectum tissue was fixed by polyformaldehyde sample fixative,and the normal paraffin section was used for immunohistochemical staining of NF-? B p65 protein.The image J software was used to quantify the average optical density of the immunohistochemical image.Results: the levels of IL-6,COX-2,TNF-? in serum and NF-? B p65 protein in anorectal tissue of model group were higher than those of normal group(P < 0.05).After 3 days of treatment,the content of IL-6,COX-2,TNF-? and the expression level of NF-? B p65 protein in anorectal tissue of the normal saline group were not different from those of the model group(P > 0.05);the content of IL-6,COX-2,TNF-? in serum and NF-? B in anorectal tissue of the combined treatment group,the fumigant group and the chuangao group were not different from those of the model group(P > 0.05)The expression level of p65 protein was lower than that of model group and normal saline group(P < 0.05);the down-regulation of serum IL-6,COX-2,TNF-? and the expression level of NF-? B p65 protein in anorectal tissue in combination group were better than that in fumigation group and hemorrhoids cream group(P < 0.05);the down-regulation of serum IL-6,TNF-? and NF-? B in anorectal tissue in fumigation group The expression level of p65 protein was better than that of Hemorrhoid Ointment group(P < 0.05).Conclusion: Zhongtong fumigant lotion can inhibit the activation of NF-? B signal pathway,decrease the content of NF-? B p65 in anorectal tissue,decrease the content of IL-6,COX-2 and TNF-? in serum,and the effect of local administration of combined with Ma Yinglong Musk Hemorrhoids cream is better.This may be related to the effect of Zhongtong fumigant lotion on reducing local inflammation after anorectal operation.
