Objective: In order to develop a great health product with the effect of improving immunity,based on the basic theory of traditional Chinese medicine,the combination and optimization of the traditional ancient prescriptions were carried out.Based on Guipi Decoction,the effective addition and subtraction were carried out by using medicine and food homologous drugs,and the Shenling Buyi Decoction was optimized.This study mainly carried out the optimization of the best extraction process and the regulation of immune activity of Shenling Buyi decoction.Method: 1.Study on the preparation process of Shenling Buyi Decoction: Investigate the factors such as decoction time,number of decoctions,and water addition multiples,and use the content of crude polysaccharide as an indicator.Orthogonal test to determine the best process.2.Kunming mice were used to establish a mouse immunosuppressive model by cyclophosphamide induction method,which was randomLy divided into 5 groups,namely,control group,model group and Decoction low,medium and high dose groups,continuous intragastric administration.30 days,then test the immune organ index,peripheral blood leukocyte count,TNF-?,IFN-? and IgG levels,spleen lymphocyte proliferation ability and apoptotic rate and mouse carbon clearance ability.3.Modulation effect of Shenling Buyi Decoction on in vitro immune function: mouse RAW264.7 macrophage cell line was used as the cell model,lipopolysaccharide LPS was used as the positive control group,cell viability was detected by CCK-8 method,and cells were measured by neutral red stain Phagocytic capacity,nitric oxide and ELISA kits were used to measure intracellular NO,TNF-?,IL-1? and IL-6 content.The mouse spleen lymphocytes were used as the cell model,and concanavalin ConA was used as the positive control group.The immunoregulatory function of Shenling Buyi Decoction was tested.The cell viability was measured by the CCK-8 method.ELISA kits were used to measure intracellular IFN-?? IL-2 content.Results: 1.Optimal decoction of Shenling Buyi Decoction: 6 medicines and 15 times water,soak 30 min in advance,decoction 3 times,each time 1.5h.2.Successfully established a mouse model of immunosuppression.The extract of Shenling Buyi Decoction can increase the viscera and thymus organ index of immunosuppressed mice caused by cyclophosphamide,increase the number of peripheral blood leukocytes,TNF-?,IgG and IFN-? in mice,and enhance lymphocyte proliferation.Viability,inhibition of the lymphocyte apoptosis rate and total apoptosis rate and enhanced mouse phagocytic function.3.Compared with the control group,Shenling Buyi Decoction can significantly promote the activity of macrophages and spleen lymphocytes without cytotoxicity,up-regulate the secretion of NO and TNF-?,IL-1? and IL-6 in macrophages and enhance phagocytic activity;Up-regulate the secretion of IFN-? and IL-2 in splenic lymphocytes.Conclusion: The best decoction process of Shenling Buyi Decoction is preferred.It was found that Shenling Buyi Decoction can comprehensively regulate the immune function of immunosuppressed mice caused by cyclophosphamide from the aspects of innate immunity and humoral immunity.Shenling Buyi Decoction can enhance the immune activity of RAW264.7 macrophages and spleen lymphocytes in mice.
