Background:Ovarian cancer is one of common gynecological cancer but has the highest mortality rate in cancers of female reproductive system.It is difficult to diagnose early and the occurence of drug resistance were the primary contributing factors.Therefore,The research on the biomarkers and therapeutic targets for ovarian cancer is one of the focuses of ovarian cancer.Objectives:To investigate the expression,clinical significance and possible mechanism of UBE2C gene in ovarian cancer to provide the theoretical base of UBE2C gene as target gene and biomarker in ovarian cancer.Methods:The ovarian cancer and normal ovarian tissues were collected from pathological department of the Second Affiliated Hospital of Kunming Medical University.The expressions of UBE2C,CCNB1 and CCNA2 in ovarian cancer and normal ovarian were examined by immunohistochemistry method,Then the relationship between the UBE2C expression and pathological characteristics were analysed.The relationship between the UBE2C expression and survival of patients with ovarian cancer using the Kaplan-Meier method to explore the prognosis significance with ovarian cancer.Then related proteins and enrichment analysis was performed to investigate the mechanism of UBE2C through genecard and STRING websits.Results:1.The expression of UBE2C in ovarian cancer was significantly higher than that in normal ovarian tissue(P<0.05).2.The expression of UBE2C gene in ovarian cancer were closely related to lymphatic metastasis,The expression of UBE2C in lymph node metastasis group was higher then that without lymph node metastasis group,there was significant difference(P<0.05).3.The expression of UBE2C gene in ovarian cancer were closely related to the degree of differentiation.The expression of UBE2C in poorly differentiation group was higher then that in moderately group and well group.the expression in moderately differentiation group was higher then that in well differentiation group,there were significant difference among them(P<0.05).4.The expression of UBE2C gene in ovarian cancer were closely related to the FIGO stage.The expression of UBE2C in ? staging was higher then that in ? and ?staging.the expression in ? staging was higher then that in ? staging,there were significant difference among them(P<0.05).5.There were no significant difference among the UBE2C gene expression and pathological classification(P>0.05).However,the expression of UBE2C in different pathological classification were all significantly higher than that in normal ovarian tissues(P<0.05).6.The expression of UBE2C is closely related to the prognosis of ovarian cancer patients.The overall survival period and disease-free survival period of the high expression group are significantly shorter than those of the low expression group(P<0.05).7.The major related proteins related to UBE2C gene included RLIM,VFLI,TCEB2,ASB2,UBE2Q2,FBXW9,UBR1,FBXL18,CUL7,FBX06,S1AH2,TRIM39,FBX09,RNF19A,BTRC,CBX4,HIST1H2AJ,HISST1H2BD,CCNB1,CCNA2,PSMD7,SHFM1,PSMD11,PSMD14,PSM1311.The Biological process of the related proteins mainly involved in the catabolic process of ubiquitin-dependent proteins,protein catabolic process,protein ubiquitination,regulation of ubiquitin-protein ligase activity in mitotic cell cycle,regulation of mitotic cell cycle and other physiological processes.8.The expression of CCNB1 and CCNA2 in ovarian cancer was significantly higher than that in normal ovarian tissue,there was significantly positive correlation between the UBE2C and CCNB1,CCNA2 genes(P<0.05).Conclusions:The expression of UBE2C gene increased in ovarian cancer and the high expression may indicate progression of disease and poor prognosis.There was significantly positive correlation between UBE2C and CCNB1,CCNA2 which suggested that CCNB1 and CCNA2 may be the downstream gene of UBE2C.However,the exactly role and mechanism need more experimental research.
