Objective:The purpose of this study was to study the effect of Danhong Injection on promoting vascular repair and inhibiting thrombosis,and to analyze the molecular mechanism of Danhong injection.Methods:A total of 30 healthy 6?8 weeks SD female rats with body weight of 250?300g were randomly divided into control group and experimental group,15 rats in each group.Five rats were killed in each group according to the sampling time(the 3rd,7th and 10th day).Construction model:free femoral artery,clamp to block the blood flow,cut off the femoral artery,wash the vascular cavity with heparin sodium physiological saline,enlarge 20 times under the microscope,use two-point suture method,use 11/0 nylon thread to anastomose the femoral artery end-to-end;after successful blood circulation,suture the wound,and cover the wound with sterile dressing.The experimental group was given 2.5 ml/kg Danhong injection every day,and the control group was given the same amount of saline every day.After 14 days of continuous feeding and regular incision dressing change,the blood samples of femoral artery were taken on the 3rd,7th and 10th day after anastomosis respectively,Platelet count and the coagulation indicators were detected,including APTT,FIB and D-dimer;the local materials were taken,the general specimens were observed to see whether the anastomotic blood vessels were unobstructed,the blood vessels adhered to the surrounding areas,swelling degree of anastomotic vessels and the healing quality of the anastomotic wound was evaluated;the number of vascular endothelial cells and fibroblasts were observed under microscope.Results:(1)Comparison of platelet number between the two groups:there was no statistical significance in the changes of platelet number in the venous blood of the two groups on the 3rd,7th and 10th day after operation.(2)Comparison of coagulation indexes between the two groups:There was no significant difference in APTT,FIB and D-dimer between the two groups.The APTT in the experimental group was significantly longer than that before the operation on the 10th day after operation,but there was no significant difference in FIB and D-dimer levels.(3)Comparison of postoperative general observation between the two groups:Thromboembolism was found in 5 anastomoses and 10 anastomoses in the experimental group and the control group,respectively.There was no significant difference between the two groups in the scores of 3 days and 7 days after operation,but the scores of the experimental group were significantly lower than those of the control group on the 10th day after operation.(4)Comparison of HE staining findings between the two groups:There was no significant difference in the number of vascular endothelial cells and fibroblasts between the experimental group and the control group on the third day after operation.The number of vascular endothelial cells and fibroblasts in the anastomotic tissue on the seventh and the tenth day after operation was higher than that in the control group.Conclusion:Danhong injection may play an important role in promoting the proliferation of vascular endothelial cells and fibroblasts after microvascular anastomosis,prolonging the anticoagulant effect of APTT and reducing thrombosis through the increase of medication time,which is of great application value in improving the quality of vascular repair,promoting and maintaining the patency of blood flow at the anastomosis.But there was no effect on FIB and D-dimer level and platelet number.
