Objective:To investigate the effect of Erdun wurile on the inflammatory response of serum and brain tissue and microglia in MCAO/R injury model rats.method:1.90 healthy male Wistar rats with body weight of 180-240g were selected.The rats were randomly divided into five groups:sham operation group,model group,low dose group,middle dose group and high dose group.The middle cerebral artery occlusion reperfusion model was established in other groups except the sham group.24 hours after the successful establishment of the model,the drug was given by gavage,0.258g/kg in the low dose group,0.516g/kg in the middle dose group and 1.032g/kg in the high dose group.The sham operation group and the model group were given pure water.The rats in the model group and each group were reperfusion for 1d,3d and 7d respectively.2.Bederson neurological function score was used to evaluate the neurological defects of rats in each group?3.TTC was used to stain the brain tissue of each group.4.Serum and brain pro-inflammatory factors IL-1??TNF-??IL-6 and anti-inflammatory factor IL-10 were detected by ELISA5.Ibal and Arg-1 were stained by immunofluorescence.result:1.The effect of Erdun wurile on the neurologic function of Bederson in MCAO/R injury model rats:the neurologic function scores of each Erdun wurile group on the first day of reperfusion were not significantly lower than those of the model group(P>0.05);the low-dose Erdun wurile group on the third day of reperfusion were not significantly lower than those of the model group(P>0.05),and the middle and high-dose Erdun wurile groups were not significantly lower than those of the model group(P>0.05)Compared with the neural function score,the model group was significantly lower,with statistical significance.The neurological score of each Erdun wurile group was significantly lower than that of the model group on the 7th day of reperfusion,which was statistically significant.2.The brain tissues of the sham group were bright red after TTC staining,and the ischemic areas of the other groups were white.3.The effects of Erdun wurile on IL-1?,TNF-?,IL-6 and IL-10 in serum and brain tissue of MCAO/R injury model rats.3.1 Compared with the model group,the levels of IL-1?,TNF-?,IL-6 in serum and brain tissue of each Erdun wurile group on the first day of reperfusion had no significant decrease(P>0.05).Compared with the model group,the levels of IL-1?,TNF-?,IL-6 in each Erdun wurile group on the third and seventh days of reperfusion were significantly lower(P<0.05).3.2 Compared with the model group,the content of anti-inflammatory factor IL-10 in serum and brain tissue of each Erdun wurile group on the first day of reperfusion had no significant increase(P>0.05),and the content of anti-inflammatory factor IL-10 in serum and brain tissue of each Erdun wurile group was significantly higher than that of the model group on the third and seventh days after administration(P?????? 0.05).4.Effects of Erdun wurile on microglia markers Ibal and Argl in MCAO/R injury model rats:4.1 Compared with the model,the Arg-1 level of each group in the first day of.reperfusion had no significant increase(P>0.05).Results:compared with the model,the three-day and seven-day reperfusion groups were significantly higher(P<0.05).4.2 no significant increase in Arg-1 was observed in the Erdun wurile group with Id reperfusion compared with the model(P>0.05).Compared with the model,the Erdun wurile group on 3d and 7d of reperfusion was significantly increased,which was statistically significant(P<0.05).Conclusion:1.Erdun wurile can improve the neurological function score of MCAO/R injury model rats.2.Erdun wurile can reduce serum and brain tissue pro-inflammatory factors IL-1?,TNF-?,IL-6,and increase anti-inflammatory factors IL-10 in MCAO/R injury model rats.3.Erdun wurile can reduce-the microglia marker Ibal in MCAO/R injury model rats,and improve the M2 microglia marker Arg-1.
