Purpose:To observe the influence of the PDS did to the SD rat brain which the MCAO caused the cerebral ischemia reperfusion injury and then adiponectin and serum soluble CD40L. To discuss the function and the understanding mechanisms of diseases that the PDS did to the cerebral ischemia reperfusion injury SD rat.Metholds:To establish a rat mode of the middle cerebral artery occlusion caused the cerebral ischemia reperfusion injury with improved Longa suture ways.To inject the PDS at enterocoelia once per day for totally seven days before doing the PDS pretreatment experiment mode. After doing the PDS treatment mode we need to inject the PDS at enterocolia once a day, totally for three days. To use HE coloration, immunohistochemical and enzyme-linked immunosorbent assay to observe the pathological alteration of brain tissue that less of blood, and detect the situation of adiponectin, serum sCD40L.Conclusion:The conclusion of pathological alteration:The pretreatment could see the cortical layer edema obviously and the space between the blood vessel getting wide. Compearing with the pretreatment mode group the edema situation of pretreatment group is better,but still more serious than the gap group and sham-operation group. The treatment group could see that the Nerve cells become edema and degeneration, the gliocyte increased and the demarcation line blurred between the grey matter at the hippocampus regions. The brain tissue of the treatment group is normal. The edema and degeneration of the nerve cells at the hippocampus rigions are better than the treatment mode group.The conclusion of Elisa experiment:Compearing with the gap group, after the rat cerebral ischemia reperfusion injury the adiponectin is obviously reduced(P<0.01)and serum sCD40L obviously increased (P<0.01) of both pretreatment mode group and treatment mode group. Comparing with two mode groups the pretreatment group the content of serum sCD40L dicreased obviously(P<0.05). The adiponectin level don’t have any diference(P>0.05). The adiponectin of treatment group was obviously increased(P<0.05) and the serum sCD40L was distinct less than treatment mode group(P<0.05).The results of immunohistochemistry:Compareing with the gap group and sham operation group, increase the content of COX-2 in brain tissue of rats in mode group Ⅰ and Ⅱ in the mode group (P<0.01); compared with the mode group, pretreatment group COX-2 decreased (P<0.05), compared with the mode group II, COX-2 in the treatment group decreased significantly (P<0.01).Conclusion:PDS can reduce the serum sCD40L level and the immiscibility of atherosclerotic plaque and reduce the plaque disruption through increasing the adiponectin level. Through the treatment of atheroma we can reducethe cerebral infarction happened.PDS can inhibit COX-2, improve the microcirculation of the brain tissue, so as to prevent the deterioration of cerebral ischemia reperfusion...
