Objective: To study the expression of PI3 K and AKT in chronic periapical periodontitis in mice by establishing a chronic periapical periodontitis model in mice,and to provide reference for further research on the role of PI3 K and AKT in chronic periapical periodontitis and the treatment of periapical disease..Methods: Twenty-five 6-week-old healthy wild-type C57 BL / 6L female mice were randomly divided into 4 experimental groups and 1 control group,with 5 in each group.The experimental group established a mouse periapical periodontitis animal model by opening the rat's bilateral mandibular first molars and exposing the medullary cavity directly to the mouth.At 1,2,3,and 4 weeks after operation,the mandibles were removed from both sides after the animals were sacrificed.The control group was mandible of healthy mice.Tissues were fixed,decalcified,embedded,and frozen sections were made.The HE staining method was used to observe the inflammation of the tissues around the apex of the mice.The TRAP staining method was used to observe the expression of osteoclasts in the inflammatory region of the tissues around the apex.Situation;The expression and distribution of PI3 K and AKT were observed by immunohistochemical staining method,and the correlation between the expression of PI3 K,AKT and osteoclast was analyzed.Results:1.HE staining results showed that there was no change in the periodontal ligament width of the periapical tissue around the healthy control group,and only few inflammatory cells were seen.One week after the operation,the periodontal ligament of the periapical tissue widened slightly.Small-scale neutrophil infiltration;2 weeks after the operation,the periodontal ligament width increased significantly,and a large number of neutrophils,macrophages,and lymphocytes infiltrated around the apical tissue,and obvious alveolar bone appeared.Absorption;The range of inflammation continued to expand at 3 and 4 weeks after pulpectomy,mainly due to lymphocyte infiltration,the alveolar bone absorption range became more pronounced,the periodontal ligament width continued to widen,and the apical periodontitis animal model was successfully established.2.TRAP staining results showed that only a few multinucleated osteoclasts were seen in the periapical tissues of the healthy control group;1 week after the operation,a small amount of osteoclasts were seen in the periapical tissues;2 weeks after the operation,the number of osteoclasts Increased;the number of osteoclasts was greater at 3 weeks after myelectomy;the number of osteoclasts decreased at 4 weeks after myelotomy.Comparing the number of osteoclasts,there was no statistical difference between 2 and 3 weeks,and no statistical difference between 1 and 4 weeks(P>0.05).There were statistical differences between the remaining groups(P<0.05).3.Immunohistochemical staining results showed that the positive cells of PI3 K and AKT were dark brown,and the expression of PI3 K and AKT showed a certain time regularity.Only a small number of PI3 K and AKT positive cells were seen in the periapical tissues of the healthy control group;positive cells increased after 1 week after myelotomy;peaked at 2 weeks after myelotomy;the number of positive cells decreased at 3 weeks;4 weeks When the number of positive cells is less.The number of positive cells in the experimental group was larger than that in the healthy control group,mainly located in neutrophils and expressed in the cytoplasm.The statistical results of PI3 K and AKT were not statistically significant between 1 week and 4 weeks(P>0.05),and the other groups were statistically significant(P<0.05).There is a moderate correlation between PI3 K OD value and AKT OD value(r=0.615,P<0.001).There is a high correlation between PI3 K OD value and osteoclast OD value(r=0.883,P<0.001).There is a high correlation between AKT OD value and osteoclast OD value(r=0.827,P<0.001).Conclusion:1.The expression of PI3 K and AKT was increased in chronic apical periodontitis.2.PI3 K and AKT participate in the disease process of chronic periapical periodontitis and have a certain relationship with bone destruction of periapical periodontitis.
