Pueraria is the dried root of the leguminous plant Puerarialobata(Willd.)Ohwi.It started in Shennong's Classic of Materia Medica,which is sweet,astringent,and cold.It mainly contains triterpenoids,isoflavones,saponins,polysaccharides and other components.It has the functions of anti-muscle and antipyretic,refreshing and quenching thirst,rash,pass through activating collaterals,detoxification and other effects,have a protective effect on liver damage.Gardenia,also known as dried jujube,is a dry,mature seed of the Hovenia dulcis Thunb plant.It started in Newly-revised Materia Medica,which is sweet and non-toxic.It mainly contains alkaloids and flavonoids,Fatty acids,saponins,glycosides,glucose and other components.It has the functions of diuretic swelling,detoxification and other effects,also has a protective effect on liver damage.Although pueraria and gardenia have been used alone or in combination with other medicinal materials to study the effects of hangover and liver protection,there has not been a match between pueraria and gardenia and the best combination.Than research data.In view of this situation,this paper has carried out research on the optimization of the extraction process of pueraria and gardenia,the preparation of pueraria granules,and the protective effect of pueraria granules on mice with chronic alcoholic liver injury.The main contents are as follows:1.The single-factor and response surface method were used to optimize the extraction process of pueraria and gardenia,and to deteramine the optimal ratio of pueraria and gardenia extracts to hangover effect,and to study the antioxidant activity of Gezhi composition in vitro.The results showed that when the ethanol concentration was 77%,the extraction temperature was 85?,the material-liquid ratio was 1:20,the extraction was performed twice,and the extraction time was 141min each time,the yield of puerarin was the highest;When the ethanol concentration was 79%,the extraction temperature was 83?,the material-liquid ratio was 1:8,the extraction was performed twice,and the extraction time was 146min each time,the yield of total flavonoids in gardenia was the highest;Based on the content of puerarin and total flavonoids,when the ratio of pueraria extract and gardenia extract is 8:2,the activity of ADH and SOD is the highest,which are 26543.04U/mL and 0.0238U/mL,indicating the combination of Gezhi composition(8:2)has the best hangover effect;The Gezhi composition(8:2)has the ability to scavenge DPPH radicals and hydroxyl radicals(IC50 values are 0.038mg/ml and 0.697mg/mL,the maximum clearance rates are 94.29%and 80.25%,respectively),with a certain degree The total reducing power(the absorbance value at the concentration of the composition of 3.6 mg/mL is 1.171),which indicates that the Gezhi composition(8:2)has strong antioxidant activity.2.Using Gezhi composition(8:2)as the main medicine,the formulation of Gezhi granules was optimized by single factor and response surface method,and the effects of Gezhi granules on ADH and AOD activities were investigated.The results showed that the comprehensive evaluation of the granules using the molding rate,dissolution rate,bulk density,angle of repose and moisture absorption rate as indicators,the best molding process for Gezhi granules is:the filler ratio is microcrystalline cellulose:dextrin 1:1.5,the main drug is added as the main drug:filler 1:1,ethanol concentration 81%.The optimal prescription of Gezhi granules is:456g of main medicine,182.4g of microcrystalline cellulose,273.6g of dextrin powder,1.14g of aspartame,1.14g of fructose,and a total of 1000g is obtained.The total value of the granules prepared with this prescription is the highest,and the preparation process is stable and reliable;Gezhi granules have uniform particles and consistent color,and the water content and solubility meet the quality standards of granules.The content of puerarin in Gezhi granules is 64.06mg/g and the total flavonoid content is 23.09mg/g.It was verified that Gezhi granules could enhance the activity of ADH and SOD,and the enzyme activity increased with the increase of the concentration of granules.3.Construction of a mouse model of chronic alcoholic liver injury.60 Kunming male mice were divided into 6 groups of 10 mice,each of which was a blank control group,a model group,and a positive control group(guilty innocence,40mg/kg).And Gezhi granules in low,medium and high dose groups(60mg/kg,120mg/kg,240mg/kg),each group was administered orally for 30min,and then 560 white wine(15mL/kg)was administered orally,and continuously administered to Wine for 45 days.The serum and liver biochemical parameters of mice were measured,and the pathological changes of liver were observed.The results showed that the ALT activity in the serum of mice at different doses of Gezhi Granules was significantly reduced,and the AST activity in the serum of mice at high doses of Gezhi Granules was significantly reduced;ADH and ALDH activities in the livers of mice in each dose group were significantly increased,MDA content was significantly reduced,and GSH content was significantly increased;SOD activity was significantly increased in the liver of Gezhi Granule high-dose group mice.In addition,Gezhi granules can improve the pathological damage of liver tissue in mice with chronic alcoholic liver injury.It shows that Gezhi granules can enhance the antioxidant capacity of mice with chronic alcoholic liver injury,inhibit lipid peroxidation in vivo,improve the pathological conditions of injured liver cells,and have a strong protective effect on mice with alcoholic liver injury.
